摘 要:目的:通过16S rDNA对比酒蒸前后黄精多糖对免疫抑制大鼠肠道菌群的影响。方法:将30只SD大鼠随机分为空白对照组、模型对照组、阳性对照组、黄精生品多糖组、黄精酒蒸品多糖组,构建免疫抑制大鼠模型,药物干预7 d后检测大鼠脏器指数、外周血细胞、免疫球蛋白补体及短链脂肪酸水平,16S rDNA测序技术对大鼠粪便肠道菌群进行基因测序。结果:与模型对照组比较,黄精生品多糖组与黄精酒蒸品多糖组均能提高免疫抑制大鼠脏器指数、外周血细胞、免疫球蛋白、补体水平及乙酸、丙酸、丁酸含量(P<0.05,P<0.01);上调Bacteroidota、Actinobacteriota、Lactobacillaceae、Lactobacillus、Ruminococcus的丰度,下调Firmicutes、Muribaculaceae、Erysipelotrichaceae、Bifidobacteriaceae、Bifidobacterium、norank_f_norank_o_Clostridia_UCG-014的丰度;且黄精酒蒸品多糖对Ruminococcaceae、Peptostreptococcaceae、Romboutsia丰度的上调作用更优。结论:黄精酒蒸品多糖提高免疫抑制大鼠免疫调节活性的作用优于黄精生品多糖,其机制可能与通过调节肠道菌群结构促进其代谢产物短链脂肪酸生成有关,该结果可为深入研究黄精酒蒸机制提供实验基础。Objective:To analysis the effects of Polygonati Rhizoma polysaccharide before and after with wine on the intestinal flora of immunosuppressed rats based on 16S rDNA.Methods:Thirty SD rats were randomly divided into blank control group,model control group,positive control group,Polygonati Rhizoma polysaccharide group and wine-steamed Polygonati Rhizoma polysaccharide group.The immunosuppressed rat model was established and were given corresponding drugs for 7 days.The organ index,peripheral blood cells,immunoglobulin complement and short-chain fatty acids levels were measured.And 16S rDNA high-throughput sequencing technology was used to perform gene sequencing on the gut microbiota of rat feces.Results:Compared with the model control group,Polygonati Rhizoma polysaccharide group and wine-steamed Polygonati Rhizoma polysaccharide group could increase the organ index,peripheral blood cells,immunoglobulin,complement,acetic acid,propionic acid and butyric acid levels of immunosuppressed rats(P<0.05,P<0.01),as well as up-regulated the abundances of Bacteroidota,Actinobacteriota,Lactobacillaceae,Lactobacillus,Ruminococcus,while down-regulated the abundances of Firmicutes,Muribaculaceae,Erysipelotrichaceae,Bifidobacteriaceae,Bifidobacterium,norank_f_norank_o_Clostridia_UCG-014.And the up-regulation effects of wine-steamed Polygonati Rhizoma polysaccharide on the abundances of Ruminococcaceae,Peptostreptococcaceae and Romboutsia was better than those of Polygonati Rhizoma polysaccharide.Conclusion:The immunomodulatory activity of wine-steamed Polygonati Rhizoma polysaccharide was better than that of Polygonati Rhizoma polysaccharide in immunosuppressed rats,and the mechanism may be related to the promotion of the production of short-chain fatty acids as metabolites by regulating the structure of intestinal flora.The results provide an experimental basis for in-depth study on the steaming with wine mechanism of Polygonati Rhizoma.
