铁皮石斛软膏对银屑病小鼠的疗效和皮肤微生物的影响

摘  要:目的:探究铁皮石斛软膏治疗银屑病的疗效以及对皮肤表面微生态的影响。方法:将12只SP F级BALB/c雄性小鼠随机分为对照组、模型组、基质软膏组、铁皮石斛软膏组,每组3只。对比各组小鼠皮损情况和银屑病面积和严重程度指数(PASI)评分,HE染色检测皮肤组织病理变化,ELISA和Western blot检测各组小鼠皮损组织中炎性因子肿瘤坏死因子α(TNF-α)、白细胞介素(IL)-1β、IL-9、IL-17含量及蛋白水平。收集各组小鼠皮损处的皮肤拭子进行16S rRNA测序分析。结果:与对照组比较,模型组和基质软膏组小鼠背部产生大量白色银屑并伴随皮肤增厚现象,且PASI评分显著升高(P<0.01),小鼠颗粒层及棘层增厚明显,棘突出现不规则延长,真皮层有明显的炎性细胞浸润现象,皮损部位炎症因子TNF-α、IL-1β、IL-9和IL-17含量和蛋白水平显著升高(P<0.01)。与模型组比较,铁皮石斛软膏干预后小鼠皮肤白色银屑明显减少,PASI评分也出现显著下调(P<0.01),HE染色观察到铁皮石斛软膏能明显改善银屑病小鼠病理状况,小鼠皮损部位炎症因子TNF-α、IL-1β、IL-9和IL-17含量和蛋白水平显著下调(P<0.01)。皮肤微生物组分析结果表明,与对照组比较,葡萄球菌属Staphylococcus在模型组和基质软膏组中丰度显著上调,乳杆菌属Lactobacillus和双歧杆菌Bifidobacterium在模型组和基质软膏组中丰度显著下调(P<0.01)。与模型组比较,铁皮石斛干预后葡萄球菌属Staphylococcus丰度显著下调,乳杆菌属Lactobacillus和双歧杆菌Bifidobacterium丰度显著上调(P<0.01)。结论:铁皮石斛软膏可能影响皮肤表面微生物组成,并减轻银屑病小鼠的炎性反应,在银屑病治疗中发挥作用。Objective:To investigate the therapeutic effect of Dendrobium candidum ointment on psoriasis mice and the effects on skin surface micro-ecology.Methods:Twelve SPF grade BALB/c male mice were randomly divided into control group,model group,matrix ointment group and Dendrobium candidum ointment group,3 in each group.The skin lesions and psoriasis area and severity index(PASI)scores of each group were compared,and the skin histopathological changes were detected by HE staining.The levels of inflammatory factors tumor necrosis factorα(TNF-α),interleukin(IL)-1β,IL-9 and IL-17 and their protein levels were detected by ELISA and Western blot.Skin swabs from the skin lesions of mice in each group were collected for 16S rRNA analysis.Results:Compared with the control group,the model group and the matrix ointment group produced a large amount of white silver dust on the back of mice accompanied by skin thickening,and the PASI score was significantly increased(P<0.01),the granular layer and the spinous layer of mice were significantly thickened,the spinous protruding irregularly extended,and the dermis showed obvious inflammatory cell infiltration.The contents and protein levels of inflammatory factors TNF-α,IL-1β,IL-9 and IL-17 were significantly increased(P<0.01).Compared with model group,Dendrobium candidum ointment significantly reduced the skin white silver dust,PASI score was also significantly decreased(P<0.01),HE staining observed Dendrobium candidum ointment can significantly improve the pathological status of psoriasis mice.The contents and protein levels of inflammatory factors TNF-α,IL-1β,IL-9 and IL-17 were significantly decreased(P<0.01).The results of skin microbiome analysis showed that compared with the control group,the abundance of Staphylococcus was significantly up-regulated in the model group and matrix ointment group,while the abundance of Lactobacillus and Bifidobacterium was significantly down-regulated in the model group and matrix ointment group(P<0.01).Compared with model group,Staphylococ
