摘 要:目的:评价多次吸入七氟烷麻醉致幼年小鼠认知功能障碍时脑神经营养因子蛋白表达情况的变化。方法:用出生第6天SPF级健康雌性C57BL/6J野生型幼鼠,采用随机数字表法分为对照组(C组)和多次吸入七氟烷麻醉组(Sev组)。Sev组于出生第6天、第8天、第10天给予3%七氟烷+60%O2,每天2 h;对照组于出生第6天、第8天、第10天在相同造模盒子内只给予60%O2,每天2 h。实验选在每天的同一时刻进行。造模结束待幼鼠呼吸平稳、完全清醒后送回母鼠笼内。造模结束后喂养幼鼠至30 d大,检测小鼠远期学习和记忆功能。于第10天幼鼠造模结束后当天取6只小鼠,深麻醉下处死后,取海马组织,采用蛋白质印迹法(Western blot)测定脑源性神经营养因子相关蛋白[cAMP反应元件(CREB)、神经营养因子3(NT-3)、神经营养因子4(NT-4)和酪氨酸激酶B(Trkb)]及海马突触后致密蛋白95(PSD95)的表达水平。结果:与C组比较,Sev组幼鼠逃避潜伏期显著延长(P<0.05),小鼠的平台穿越次数减少(P<0.05)。与C组比较Sev组幼鼠血清炎症因子表达增高(P<0.05),海马组织中脑源性神经营养因子相关蛋白(CREB,BDNF,NT-3,NT-4和Trkb)及PSD95表达降低(P<0.05)。结论:多次吸入七氟烷麻醉致幼年小鼠认知功能障碍可能与脑神经营养因子相关蛋白(CREB,BDNF,NT-3,NT-4和Trkb)及PSD95的表达降低相关。Objective:To evaluate the changes in the expression of neurotrophin protein in the brain of young mice with cognitive dysfunction induced by repeated inhalation of sevoflurane anesthesia.Methods:SPF healthy female C57BL/6J young mice at the birth of 6 days were divided into 2 groups using a random number table method:control group(C group)and multiple inhalations of sevoflurane anesthesia group(Sev group).Sev Group was given 3%sevoflurane+60%O 2,2 h a day for 3 days.The control group was only given 60%O 2 in the same modelling box,2 h a day interrupted for 3 days,and the experiment was carried out at the same time every day.At the end of modelling,the young rats were returned to the mother rat cage after breathing was stable and completely awake.After modelling,the young mice were fed for 30 days,and the long-term learning and memory function of the mice was detected.At the end of modelling,6 mice were sacrificed after deep anesthesia,and the hippocampus was obtained for determination of the expression of brain-derived neurotrophic factor-related proteins[cAMP reaction element(CREB),Neurotrophin 3(NT-3),Neurotrophin 4(NT-4)and tyrosine kinase B(Trkb)]and synaptic plasticity protein(PSD95)(by Western blot).Results:Compared with the C group,the escape latency of the young mice in the Sev group was significantly prolonged(P<0.05),and the number of platform crossings was reduced(P<0.05).The results of enzyme-linked immunosorbent assay(ELISA)showed the expression of inflammatory factors in serum was increased in the Sev group(P<0.05).The expression of brain-derived neurotrophic factor proteins(CREB,BDNF,NT-3,NT-4 and Trkb)and synaptic plastic-related protein PSD95 in the hippocampus of young mice in the Sev group were decreased(P<0.05).Conclusion:The cognitive impairment induced by repeated inhalation of sevoflurane anesthesia in young mice may be related to the decreased expression of brain neurotrophic factor-related proteins(CREB,BDNF,NT-3,NT-4 and Trkb proteins)and hippocampal synaptic plasticity-related protein P
