摘  要:目的分析包头市布鲁菌的分子流行病学特征,为控制人间布鲁菌病提供科学依据。方法11株布鲁菌来源于包头市2012-2022年布鲁菌病患者血液培养物。采用多位点序列分型(MLST)技术分析菌株序列型,采用核心基因组单核苷酸多态性(cgSNP)分析方法,了解包头市菌株与国内菌株间的遗传进化关系。结果包头市的11株羊种布鲁菌经MLST分型方法鉴定为ST8型(3-2-3-2-1-5-3-2-8),经cgSNP分析并进行系统地理重建,所研究的菌株属于东地中海谱系(Ⅱ基因型),与我国其他9个地区的52株羊种布鲁菌共检测到4236个SNP,被聚为5个群,与内蒙古自治区乌兰察布市、黑龙江省哈尔滨市和新疆维吾尔自治区的羊种布鲁菌亲缘关系密切。结论包头市布鲁菌病疫情的病原体可能存在多条传播链。cgSNP作为传统生物学鉴定补充技术方法,可用于布鲁菌遗传多样性分析和分子流行病学溯源调查,以提高布鲁菌病监测能力。Objective To study the molecular epidemiological characteristics of Brucella in Baotou city,and provide scientific basis for the control of human brucellosis.Methods A total of 11 Brucella strains were obtained from blood cultures of patients with brucellosis from 2012 to 2022.Multilocus sequence typing(MLST)was used to analyze the sequence type of the strains.Core genome single nucleotide polymorphism(cgSNP)analysis was used to analyze the genetic evolution relationship between Baotou strains and domestic strains.Results A total of 11 strains of Brucella melitensis isolated from Baotou city were identified as ST8 type(3-2-3-2-1-5-3-2-8)by MLST.cgSNP analysis and phylogeographic reconstruction showed that the studied strains belonged to the Eastern Mediterranean lineage(genotypeⅡ).A total of 4236 SNPS were detected with 52 strains of B.melitensis isolated from 9 other regions.The isolates were grouped into five clusters,which were closely related to the isolates from Ulanqab of Inner Mongolia Autonomous Region,Harbin of Heilongjiang Province and Xinjiang Uygur Autonomous Region.Conclusions There might be multiple transmission chains of the pathogen of brucellosis in Baotou city.cgSNP as a complementary technical method to traditional biological identification,could be used to analyze the genetic diversity of Brucella and to investigate the molecular epidemiology of brucellosis,so as to improve the ability of brucellosis surveillance.
