芋蝗线粒体全基因组特征及稻蝗亚科线粒体基因组比较分析

摘  要:芋蝗(Gesonula punctifrons)是莲藕和芋类等水生蔬菜的一种重要害虫,为探究其线粒体基因组结构及稻蝗亚科(Oxyinae)的线粒体基因组进化特征,本研究利用二代测序技术获取芋蝗线粒体全基因组序列,并对芋蝗和其他已报道的11个稻蝗亚科物种的线粒体基因组的基因结构特点及碱基组成等进行比较分析。芋蝗线粒体基因组全长为16312 bp,A+T含量76.16%,共包含13个蛋白编码基因(protein-coding genes,PCGs)、22个转运RNA(transfer RNA,tRNAs)基因、2个核糖体RNA(ribosomal RNAs,rRNAs)基因和1个长度为1496 bp的AT富集区(AT-rich region)。稻蝗亚科物种的线粒体基因组长度在15427~16402 bp之间,其全基因组序列均呈现明显的AT偏好性(74.75%~76.16%)。tRNA基因二级结构分析显示,除tRNA^(Ser(AGN))缺少DHU臂而形成简单的环,其余tRNA基因均能形成典型的三叶草结构。基于13个PCGs,采用贝叶斯法(Bayesian inference method,BI)和最大似然法(maximum likelihood method,ML)重建稻蝗亚科系统发育树,结果显示芋蝗位于稻蝗亚科的最基部。本研究获得了芋蝗线粒体基因组全序列,对稻蝗亚科进行线粒体基因组学比较分析,研究结果不仅能够为直翅目(Orthoptera)分子系统学研究提供基础数据,更有助于阐述稻蝗亚科线粒体基因组分化规律。Gesonula punctifrons is one of important pests affecting aquatic vegetables such as lotus roots and taros.To investigate its mitogenome structure and the evolutionary feature of the mitogenomes among the subfamily Oxyinae,this study obtained the complete mitogenome sequence of Gesonula punctifrons using second-generation sequencing technology.Comparative analysis were conducted on the mitogenome structure and base composition of G.punctifrons and 11 other reported Oxyinae species.The mitogenome of Gesonula punctifrons was 16312 bp in size with an A+T content of 76.16%.It included 13 protein-coding genes(PCGs),22 transfer RNAs(tRNAs)genes,2 ribosomal RNAs(rRNAs)genes,and an AT-rich region.The mitogenome sizes of Oxyinae species ranged from 15427 to 16402 bp,with all sequences exhibited a distinct A+T bias(74.75%~76.16%).Secondary structure analyses of tRNAs revealed that apart from tRNA^(Ser(AGN))lacking a DHU arm and forming a simple loop,the remaining genes could form a typical cloverleaf structure.Bayesian inference(BI)and maximum likelihood(ML)methods were employed to reconstruct the phylogenetic trees of Oxyinae using 13 PCGs.The results showed that Gesonula punctifrons occupied the basal position of Oxyinae.This study provided the mitogenome sequence of Gesonula punctifrons and conducted comparative mitochondrial genomics analysis of Oxyinae.These findings not only contribute foundational data for molecular systematic studies in Orthoptera,but also aid in elucidating the differentiation patterns of Oxyinae mitogenomes.
