摘  要:目的:探讨木犀草素对人卵巢癌A2780细胞的作用,并揭示其可能机制。方法:通过CCK-8检测木犀草素对A2780细胞活力的影响,确定加药浓度;克隆斑、划痕和Transwell实验检测木犀草素对A2780细胞增殖和转移能力的影响;并使用流式细胞术检测木犀草素作用下细胞凋亡水平的变化;凋亡、自噬及AKT-mTOR信号通路相关蛋白的表达情况则通过Western blot实验检测。结果:木犀草素抑制了A2780细胞活力、增殖和转移的能力;流式细胞术结果表明木犀草素呈浓度依赖性诱导A2780细胞凋亡;且自噬相关蛋白LC3-Ⅱ/LC3-Ⅰ比例增加,P62表达下降,P-AKT、P-mTOR与抗凋亡蛋白Bcl-2表达水平降低。结论:木犀草素通过AKT-mTOR信号通路诱导人卵巢癌A2780细胞凋亡和自噬,从而抑制卵巢癌细胞恶性进展。Objective:To investigate the effect of luteolin on human ovarian cancer A2780 cells and reveal its possible mechanism.Method:The effect of luteolin on the viability of A2780 cells was detected by CCK-8,and the concentration of luteolin was determined;the effects of luteolin on the proliferation and metastasis of A2780 cells were detected by clone spot,scratch,and Transwell assay;flow cytometry was used to detect the changes of apoptosis level under the action of luteolin;the expression of proteins related to apoptosis,autophagy,and AKT-mTOR signaling pathway was detected through Western blot experiments.Re⁃sults:Luteolin inhibited the viability,proliferation,and metastasis of A2780 cells;the proportion of autophagy related protein LC3-Ⅱ/LC3-Ⅰincreased,the expression of P62 decreased,and the expression levels of P-AKT,P-mTOR,and anti-apoptotic protein Bcl-2 decreased.Conclusion:Luteolin induces apoptosis and autophagy in human ovarian cancer A2780 cells through AKTmTOR signaling pathway,thereby inhibiting the malignant progression of ovarian cancer cells.
