摘 要:为探究红花药材中多糖的最佳提取工艺,以新疆塔城地区的红花药材为原料,采用水提醇沉法从红花药材中提取多糖,并进行初步纯化。通过查阅文献,选取时间、料液比、沉淀试剂浓度、温度四个影响较大的因素进行单因素实验,然后分别从四个单因素中选择三个影响较大的水平,进行四因素三水平的正交实验确定最佳提取工艺。结果表明,最佳提取工艺为:时间2 h/次(共三次)、料液比1∶20、温度90℃、沉淀试剂浓度95%乙醇,影响因素中料液比>温度>时间>沉淀试剂浓度,最后得到的粗多糖纯度为12.64%,经反复冻融除杂、Sevage法除蛋白、H_(2)O_(2)脱色后多糖纯度达80.1%。To explore the optimal extraction process for polysaccharides in safflower medicinal materials,water extraction and alcohol precipitation method was used to extract polysaccharides from safflower medicinal materials in Tacheng,Xinjiang,and preliminary purification was carried out.By consulting literature,four factors with significant effects,namely time,solid-liquid ratio,precipitation reagent concentration and temperature,were selected for single factor experiments.Then,three levels with significant effects were selected from each of the four single factors,and orthogonal experiments with four factors and three levels were conducted to determine the optimal extraction process.The results showed that the optimal extraction process was as follows:with 2 h/time(three times in total),solid-liquid ratio was 1∶20,temperature was 90℃,and precipitation reagent concentration was 95%ethanol.The influencing factors were as follows:solid-liquid ratio>temperature>time>precipitation reagent concentration.The final crude polysaccharide purity was 12.64%.After repeated freeze-thaw impurity removal,Sevage protein removal and H_(2)O_(2) decolorization,the polysaccharide purity reached 80.1%.
