摘 要:为了解光唇鱼Acrossocheilus fasciatus自然分布的不同地理群体遗传多样性和遗传结构,基于cyt b基因序列,对浙江省的苕溪、金华江、曹娥江、椒江和安徽省的新安江、青弋江共6个水系的116尾光唇鱼进行分析。结果显示,116条cyt b基因序列定义了12个单倍型,光唇鱼6个群体的单倍型多样性为0~0.648,核苷酸多样性为0~0.004 35,除曹娥江群体呈现高单倍型多样性和低核苷酸多样性的特点外,其他群体均呈现出低单倍型多样性和低核苷酸多样性的特点。遗传分化指数分析显示,除苕溪与椒江群体间属中度遗传分化外,其余各群体间均属于极大的遗传分化(P<0.01)。基于单倍型序列构建的系统发育树显示,新安江、金华江、曹娥江、苕溪和椒江5个群体的单倍型混杂在一起聚为一支,青弋江群体不同单倍型聚为一支。遗传距离分析结果显示,青弋江群体与其他群体间的遗传距离均大于0.03。综合遗传距离、遗传分化指数和单倍型系统发育树结果,青弋江群体与其他群体间产生了明显的遗传分化。核苷酸错配分布和中性检验结果显示,光唇鱼近期并未经历种群扩张事件。本研究结果为光唇鱼种质资源保护与利用、增殖放流和良种选育等相关工作提供了理论依据。Based on cyt b sequence,the genetic diversity and genetic structure of 116 individuals of Acrossocheilus fascia-tus from 6 geographical populations in Tiaoxi,Jinhuajiang,Caoejiang and Jiaojiang of Zhejiang Province and Xin’anjiang,Qingyijiang of Anhui Province were analyzed.The results showed that 116 cyt b sequences defined 12 haplotypes,and the haplotype diversity and nucleotide diversity of the 6 populations ranged from 0 to 0.648 and from 0 to 0.00435,respec-tively.Caoejiang population showed high haplotype diversity and low nucleotide diversity,while the others showed low hap-lotype diversity and low nucleotide diversity.The analysis of genetic differentiation index showed that the genetic differen-tiation was moderate between Tiaoxi and Jiaojiang populations,while the genetic differentiation was very high among the other populations(P<0.01).The phylogenetic tree constructed with the haplotype sequences showed that,the haplotypes of Xin’anjiang,Jinhuajiang,Caoejiang,Tiaoxi and Jiaojiang populations were clustered into one clade,while the different haplotypes of Qingyijiang population were distributed in another.The genetic distance analysis showed that the genetic dis-tance value between Qingyijiang population and other populations was larger than 0.03.The combined results from analy-sis of genetic distance,genetic differentiation index and haplotype phylogenetic tree confirmed the obvious genetic differen-tiation between the Qingyijiang population and other populations.The results of nucleotide unmatched distribution and neu-trality tests showed that the species of A.fasciatus had not experienced population expansion events recently.Therefore,this study provides theoretical basis for the conservation and utilization of germplasm resources,stock enhancement,and ge-netic breeding of A.fasciatus.
