摘  要:单胺氧化酶A(monoamine oxidase A,MAOA)是一种膜结合线粒体酶,几乎存在于所有脊椎动物组织中,催化生物源性单胺和膳食衍生单胺的降解。MAOA主要调节神经递质代谢,但与抗肿瘤免疫反应密切相关。以往研究更多关注MAOA在肿瘤细胞中的作用,新近研究发现,MAOA在肿瘤相关免疫细胞中同样发挥重要作用。本文综述了MAOA对抑制性肿瘤微环境(tumor microenvironment,TME)的调节作用,以及通过直接影响免疫细胞内单胺类物质及其代谢,进而对各类肿瘤相关免疫细胞(如CD8+T细胞和肿瘤相关巨噬细胞)发挥的抑制作用;阐明开发新型MAOA抑制剂药物和多药联合可能提高肿瘤免疫治疗效果,提出MAOA可能作为1个肿瘤免疫新靶点,影响肿瘤免疫治疗效果。Monoamine oxidase A(MAOA)is a membrane-bound mitochondrial enzyme that exists in almost all vertebrate tissues,where it catalyzes the degradation of biogenic and dietary-derived monoamines.MAOA has the function of regulating neurotransmitter metabolism and is associated with anti-tumor immune responses.Most previous studies have focused on the role of MAOA in tumor cells,while more recent findings suggest that MAOA plays an equally significant role in tumor-associated immune cells.In this review,we summarize the regulatory effect of MAOA on the inhibitory tumor microenvironment.The suppressing function of MAOA on various types of tumor-associated immune cells(e.g.,CD8+T cells and tumor-associated macrophages)by its direct effect on monoamines and their metabolic characteristics are discussed.We propose that developing novel MAOA-inhibitor drugs and exploring multidrug-combination strategies may enhance the efficacy of immune therapy for tumors.In conclusion,MAOA may act as a novel target in tumor immunity and influence the effect of tumor immunotherapy.
