摘 要:为了探究低血清培养基对Marc-145细胞增殖与产毒能力的影响,本试验在低血清培养基加入不同体积分数的牛血清后,分别在方瓶与转瓶上培养Marc-145细胞,培养3代后进行接毒、收获抗原。检测每次传代后培养48 h的细胞密度与抗原效价,培养基添加3%~4%血清,细胞密度与对照组较一致,同时低血清培养基培养的Marc-145细胞对PRRSV的产毒能力低于对照组约0.2~0.3 lgTCID50/mL。结果表明:低血清培养基培养Marc-145细胞可将血清添加量降低至3%~4%,产毒能力符合产品质量要求,该数据可为Marc-145细胞低血清或无血清培养提供参考。In order to investigate the effect of low serum culture medium on the proliferation of Marc-145 cells and virus,this experiment added different volume fractions of bovine serum to low serum culture medium,and cultured Marc-145 cells on different volume culture flasks,respectively.After 3 passages of cultivation,the cells were inoculated and antigens were harvested.The cell density and antigen titer of Marc-145 cells cultured for 48 hours after each passage were tested.With the serum addition of 3%to 4%,the cell density was more consistent with the control group by using low serum medium,and the antigen titer of Marc-145 cells cultured with simultaneous low serum medium to PRRSV was approximately 0.2 to 0.3 lgTCID50/mL lower than the control group.The results showed that the culture of Marc-145 cells in low serum culture medium could reduce the amount of serum addition to 3%~4%,and the antigen production capacity meets the product quality requirements.This data can provide a reference for the culture of Marc-145 cells with low serum or no serum.
