摘 要:以水草红丝青叶组培苗为试验材料,研究不同浓度生长素(IAA、IBA、2,4-D)和6-KT在组培培养基中单独或联合使用对其生长及生理应答的变化影响。结果表明,在0.5~3.0μg/μL范围内,单独使用IBA或2,4-D的组培苗表型优于单独使用IAA或6-KT。当IAA或6-KT浓度等于或大于2.0μg/μL时,组培苗发育迟缓;在1.0μg/μL IBA和1.0μg/μL 6-KT或0.5μg/μL IBA和2.0μg/μL 6-KT联合使用早期,组培苗长势均突出,叶片的丙二醛含量较低,叶绿素、可溶性蛋白和脯氨酸含量较高。1.0~2.0μg/μL IBA和2.0μg/μL 6-KT联合使用早期抑制组培苗生长,长期使用会导致其茎尖烧灼。The tissue culture seedling of water plant Hygrophila polysperma was taken as the experimental material,the effects of different concentrations of auxin(IAA,IBA,2,4-D)and 6-KT on the growth and physiological response were studied in tissue culture medium.The results showed that the phenotype of tissue culture seedlings using IBA or 2,4-D alone was superior to IAA or 6-KT in the range of 0.5-3.0μg/μL.When IAA or 6-KT concentration was equal to or greater than 2.0μg/μL,the growth of tissue culture seedlings was delayed.At the early stage of combined use of 1.0μg/μL IBA and 1.0μg/μL 6-KT or 0.5μg/μL IBA and 2.0μg/μL 6-KT,the growth of tissue culture seedlings was prominent,and the contents of malondialdehyde in leaves were lower,and the contents of chlorophyll,soluble protein and proline were higher.The combination of 1.0-2.0μg/μL IBA and 2.0μg/μL 6-KT inhibited the growth of tissue culture seedlings in the early stage,and led to the burning of the stem tips in the long term.
