云南栘[木衣]离体快繁和再生体系的建立

摘  要:为建立云南栘[木衣]高效离体快繁体系,本研究以云南栘[木衣]带腋芽茎段和叶片为外植体,围绕外植体消毒、抑制外植体褐化、初代与继代培养、壮苗与生根培养以及愈伤组织的诱导等进行研究。结果表明,茎段初代培养的最适培养基为:MS+6-BA 3 mg/L+NAA 0.2 mg/L;继代培养最适培养基为:MS+6-BA 4 mg/L+NAA 0.3 mg/L;壮苗配方为MS+6-BA 2 mg/L+NAA 0.3 mg/L时组培苗生长高大,为MS+6-BA 1 mg/L+NAA 0.3 mg/L时组培苗生长粗壮;生根培养最适培养基为:1/2 MS+IBA 2.0 mg/L。叶片愈伤组织诱导最佳培养基为:MS+NAA 0.6 mg/L+TDZ 3 mg/L,在MS+6-BA 3 mg/L+IBA 0.2 mg/L培养基中能直接诱导叶片产生不定芽。In order to establish an efficient in vitro rapid propagation system of Docynia delavayi(Franch.)Schneid.from Yunnan Province,the stems and leaves of Docynia delavayi(Franch.)Schneid.with axillary buds were used as explants.The disinfection of explants,inhibition of explants browning,primary and secondary culture,strong seedling and rooting culture,and callus induction were studied.The results showed that the optimal medium for stem segment primary culture was MS+6-BA 3 mg/L+NAA 0.2 mg/L.The optimal medium for subculture was MS+6-BA 4 mg/L+NAA 0.3 mg/L.When the formula of strong seedling was MS+6-BA 2 mg/L+NAA 0.3 mg/L,the tissue culture seedling grew tall,and when MS+6-BA 1 mg/L+NAA 0.3 mg/L,the tissue culture seedling grew robust.The optimal rooting culture was 1/2 MS+IBA 2.0 mg/L.The optimal medium for leaf callus induction was MS+NAA 0.6 mg/L+TDZ 3 mg/L,and MS+6-BA 3 mg/L+IBA 0.2 mg/L could directly induce adventitious buds in leaves.
