摘 要:制备透明质酸脂质体并对其进行表征和透皮性能的研究。采用薄膜分散法制备透明质酸脂质体,透射电子显微镜(TEM)观察形态,马尔文粒度仪测定粒径和电位。使用TK-6H1型透皮扩散试验仪研究脂质体的体外透皮性能,并评估了透明质酸脂质体和透明质酸溶液的体外经皮渗透行为差异,还对透明质酸抗氧化能力、透明质酸脂质体明目效果进行研究。结果表明,透明质酸脂质体呈类球形囊泡结构,粒径为(90.70±1.20)nm,多分散性指数(PDI)为(0.241±0.021),Zeta电位为(-40.65±0.781)mV。透明质酸脂质体的8 h累积经皮渗透量是透明质酸溶液的5倍。此外,通过DPPH自由基清除实验和脂多糖刺激实验,证明透明质酸具有一定的抗氧化及明目效果,且无明显细胞毒性。脂质体能显著促进透明质酸的透皮吸收,有望用于天然生物大分子的经皮递送系统。Preparation of hyaluronic acid liposomes and characterization of their transdermal properties.Hyaluronic acid liposomes were prepared by thin film dispersion method,and the morphology was observed by transmission electron microscopy(TEM).The particle size and potential were measured by Malvern particle size analyzer.The in vitro transdermal performance of liposomes was studied using the TK-6H1 transdermal diffusion tester,and the differences in in in vitro transdermal penetration behavior between hyaluronic acid liposomes and hyaluronic acid solutions were evaluated.In addition,the antioxidant capacity of hyaluronic acid and the visual acuity of hyaluronic acid liposomes were also studied.The results showed that hyaluronic acid liposomes had a spherical vesicular structure with a particle size of(90.70±1.20)nm,a polydispersity index(PDI)of(0.241±0.021),and a Zeta potential of(-40.65±0.781)mV.The 8-hour cumulative transdermal permeability of hyaluronic acid liposomes is 5 times that of hyaluronic acid solution.In addition,through DPPH radical scavenging experiments and lipopolysaccharide stimulation experiments,it has been demonstrated that hyaluronic acid has certain antioxidant and vision improving effects,and has no significant cytotoxicity.Liposomes can significantly promote the transdermal absorption of hyaluronic acid and are expected to be used in the transdermal delivery system of natural biomolecules.
