摘  要:目的:分析多肽水凝胶负载人尿源性干细胞移植治疗创伤性脑损伤大鼠的可行性。方法:选取2022年1月—2024年1月南昌大学第一附属医院神经外科购入的40只Wistar大鼠,制备成创伤性脑损伤大鼠,采用随机数字表法分为对照组和观察组,各20只。对照组进行人尿源性干细胞移植治疗,观察组进行多肽水凝胶负载人尿源性干细胞移植治疗。比较两组移植细胞脑内存活和分化能力、移植细胞巢蛋白(Nestin)、胶质纤维酸性蛋白(GFAP)、β微管蛋白Ⅲ(β-tubulinⅢ)表达水平。结果:观察组增强绿色荧光蛋白与红色荧光蛋白体积大于对照组。观察组Nestin、GFAP、β-tubulinⅢ表达水平均高于对照组(P<0.05)。结论:多肽水凝胶负载人尿源性干细胞移植治疗创伤性脑损伤大鼠的效果更佳,且人尿源性干细胞便于采集、定向移植优势明显。Objective:To analyze the feasibility of the transplantation of human urine-derived stem cells loaded with polypeptide hydrogel in treatment of traumatic brain injury rats.Methods:Forty Wistar rats purchased by the Neurosurgery Department of the First Affiliated Hospital of Nanchang University from January 2022 to January 2024 were selected to prepare traumatic brain injury rats.These rats were divided into control group and observation group according to random number table method,with 20 rats in each group.The control group received human urine-derived stem cell transplantation,while the observation group received human urine-derived stem cell transplantation loaded with polypeptide hydrogel.The survival and differentiation abilities of transplanted cells in the brain,as well as the expression levels of Nestin,glial fibrillary acidic protein(GFAP),and β-tubulin Ⅲ in transplanted cells,were compared between the two groups.Results:The volumes of enhanced green fluorescent protein and red fluorescent protein in the observations were greater than those in the control group.The expression levels of Nestin,GFAP and β-tubulin Ⅲ in the observation group were higher than those in the control group(P<0.05).Conclusion:The transplantation of human urine-derived stem cells loaded with polypeptide hydrogel in the treatment of traumatic brain injury rats has better effects,and human urine-derived stem cells have obvious advantages in easy collection and directional transplantation.
