钩吻素子对小鼠急性肺损伤的治疗作用

摘　　要：目的探讨钩吻素子对小鼠急性肺损伤的治疗作用,为钩吻素子的临床应用提供理论依据。方法除空白组外,造模成功的ICR雄性小鼠随机分成模型组、钩吻素子组、地塞米松磷酸钠组,药物治疗后4 h和72 h分批处死小鼠。然后测定血液中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素1(Interleukin-1β,IL-1β)和白细胞介素6(Interleukin-6,IL-6)水平,计算肺系数,观察肺组织病理学变化,以及检测肺组织MPO、MDA含量和SOD活力。结果钩吻素子能抑制小鼠血液中TNF-α、IL-1β和IL-6生成与释放,降低肺组织系数,改善肺脏组织病理学形态并减少肺组织MPO、MDA含量以及提高SOD活力。结论钩吻素子对小鼠急性肺损伤具有治疗作用,可能是通过抑制炎症细胞因子生成,提高机体的抗氧化能力等产生治疗作用。Objective To investigate the therapeutic effect of koumine on acute lung injury in mice,and to provide theoretical basis for clinical application of koumine.Methods Except for the control group,successfully modeled ICR male mice were randomly divided into model group,koumine group,and dexamethasone(DEX)group.The mice were sacrificed at 4 h and 72 h after treatment,respectively.Then the levels of tumor necrosis factor-α(TNF-α),Interleukin-1β(IL-1β),Interleukin-6(IL-6)in blood were determined,the lung coefficient were calculated,the pathological changes of lung tissue were observed and the contents of MPO,MDA and the activity of SOD in lung tissue were measured.Results The results showed that the production and release of TNF-α,IL-1βand IL-6 in peripheral blood were inhibited by koumine.Koumine can also reduce the lung tissue coefficient,improve the histopathological morphology of lung tissues and decrease the contents of MPO and MDA,increase the activity of SOD.Conclusion Koumine has a therapeutic effect on acute pleurisy in mice,which may be through inhibiting the production of inflammatory cytokines and improving the antioxidant capacity of the body.
