铁皮石斛提取物对急性酒精性胃溃疡小鼠的保护作用

摘　　要：目的研究铁皮石斛提取物对急性酒精性胃溃疡小鼠的作用,并初步探讨其改善胃溃疡的潜在作用机制。方法将60只雄性C57BL/6小鼠随机分为正常对照组,模型组,铁皮石斛提取物低、中、高剂量组和奥美拉唑(omeprazole,OMZ)组。预灌胃给药7 d后,模型组和各给药组灌胃体积分数90%酒精建立急性胃溃疡模型。测定各组小鼠胃溃疡损伤指数、溃疡抑制率;采用酶联免疫吸附法(enzyme linked immunosorbent assay,ELISA)检测血清白介素-6(interlukin-6,IL-6)、肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)、前列腺素2(prostaglandin E2,PGE2)、环氧合酶1(cyclooxygenase 1,COX-1)、超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)、还原型谷胱甘肽(glutathione,GSH)及过氧化氢酶(micro catalase,CAT)的含量;HE染色法观察胃溃疡组织病理学变化;采用免疫组织化学方法考察各组小鼠胃黏膜表皮生长因子(epidermal growth factor,EGF)蛋白的表达情况。结果铁皮石斛提取物和奥美拉唑能改善酒精性胃溃疡的病理学变化。与模型组相比,铁皮石斛提取物能显著提高SOD、CAT、GSH的活力及增强PGE2、COX-1的水平,降低MDA的含量及血清IL-6和TNF-α的水平,并增加胃黏膜组织EGF的阳性细胞表达。结论铁皮石斛提取物对酒精性胃溃疡具有一定的预防保护作用,其机制可能与抑制炎症反应和氧化应激,增强胃黏膜保护因子的防御作用有关。Objective To study the effect of Dendrobium officinale extract on acute alcoholic gastric ulcer in mice,and to explore the potential mechanism of improving gastric ulcer.Methods 60 male C57BL/6 mice were randomly divided into normal control group,model group,low-dose,middle-dose,high-dose extract of Dendrobium officinale group and omeprazole group.After pre-administration of Dendrobium officinale extract and omeprazole for seven days,the model of acute gastric ulcer was established by 90%ethanol in model group and medication group.The gastric ulcer injury index and ulcer inhibition rate were determined.Serum levels of interleukin-6(IL-6),tumor necrosis factorsα(TNF-α),prostaglandin E 2(PGE 2),cyclooxygenase 1(COX-1),superoxide dismutase(SOD),malondialdehyde(MDA),glutathione(GSH)and micro catalase(CAT)were measured by enzyme linked immunosorbent assay(ELISA).And pathological sections of gastric tissues were observed by hematoxylin-eosin staining.The expression of gastric epidermal growth factor(EGF)protein was examined by immunohistochemistry.Results Dendr obium officinale extract and omeprazole could improve the pathological changes of ethanol-induced gastric ulcer.Compared with the model group,Dendrobium officinale extract significantly increased the activities of SOD,CAT,GSH and enhanced the levels of PGE2 and COX-1,decreased the content of MDA and the levels of serum IL-6 and TNF-α,and increased the positive cell expression of EGF in gastric mucosal tissue.Conclusion Dendrobium officinale extract has a protective effect on alcoholic gastric ulcer,which may be related to the inhibition of inflammatory reaction and oxidative stress,and the enhancement of gastric mucosal protective factors.
