人参归脾丸防治环磷酰胺化疗胃肠道反应的实验研究

摘　　要：目的探究人参归脾丸对环磷酰胺致胃肠道反应小鼠的干预作用。方法选取60只SPF级BALB/c小鼠随机分为阴性对照组,模型组,盐酸昂丹司琼片组[2.4 mg/(kg·d)]及人参归脾丸高剂量组[5.4 g/(kg·d)]、中剂量组[2.7 g/(kg·d)]、低剂量组[1.35 g/(kg·d)],每组10只。除阴性对照组外其余5组动物分别以80 mg·kg^(-1)剂量的环磷酰胺进行隔日腹腔注射,共注射3次,造模同时连续灌胃给予相应药物6 d,每日观察动物状态。隔日称量各组小鼠造模后的体重,给药结束检测胃液pH值,计数小鼠肠道派氏结(PP结),免疫组化法检测小鼠胃肠道组织中TLR4蛋白的表达。结果人参归脾丸能够改善环磷酰胺造模小鼠精神状态,促进模型小鼠体重增长,增加PP结数量,提高胃液pH值,下调胃、十二指肠、空肠及回肠组织TLR4蛋白的相对表达量。结论人参归脾丸对环磷酰胺化疗所致胃肠道反应有一定的预防治疗作用,其机制可能与调节胃肠道TLR4蛋白表达有关。Objective To determine the intervention effect of Renshen Guipi pills on mice with cyclophosphamide-induced gastrointestinal reaction.Methods Totally 60 SPF BALB/c mice were randomly divided into 6 groups:a negative control group,a model group,an Ondansetron hydrochloride tablet group[2.4 mg/(kg·d)],and Renshen Guipi pills high[5.4 g/(kg·d)],medium[2.7 g/(kg·d)]and low[1.35 g/(kg·d)]dose groups,with 10 mice in each group.Except for the negative control group,the other 5 groups were intraperitoneally given 80 mg·kg^(-1) of cyclophosphamide once every other day,3 injections in total.At the same time,the mice were continuously intragastrically gavaged with corresponding drugs for 6 days,and their daily condition was observed.The weight,pH value of gastric juice,and small intestinal PP nodes of mice were measured after the modeling.Immunohistochemistry was used to detect the expression of TLR4 protein in the gastrointestinal tract of the mice.Results Renshen Guipi pills improved the mental state of cyclophosphamide model mice,promoted the body weight growth of model mice,increased the pH value of gastric juice and the number of PP nodes in the small intestine,and down-regulated the relative expression of TLR4 protein in the gastric,duodenum,jejunum and ileal tissues.Conclusion Renshen Guipi pills prevent and treat gastrointestinal reaction induced by cyclophosphamide chemotherapy,whose mechanism may be related to the regulation of gastrointestinal TLR4 protein expression.
