摘 要:目的探讨食消饮对食积小鼠的药效作用。方法将120只ICR小鼠随机均分为两个实验(实验A和B),每个实验的小鼠均分为正常组,模型组,食消饮低、中、高剂量组和健胃消食片组。食积模型构建后,各给药组灌胃相应受试药物,每日2次,连续7 d。其中实验A小鼠用于脏器系数、血清生化指标、小肠免疫组化、结肠HE染色以及肠道菌群的检测,实验B小鼠用于胃排空率和小肠推进率实验。结果与模型组相比,食消饮高剂量组小鼠脾脏脏器系数明显升高,中、高剂量组小鼠血清生化指标均得到明显改善,小肠推进率升高、胃内残留率降低。免疫组化结果显示,食积模型小鼠小肠中C-kit表达明显降低,食消饮中、高剂量和健胃消食片组小鼠中C-kit表达明显升高。HE结果显示,食消饮中、高剂量组小鼠结肠绒毛长度均增加、隐窝深度均降低、绒毛长度/隐窝深度的比值增加。粪便菌群计数结果显示,食消饮高剂量组小鼠粪便中乳杆菌和双歧杆菌增加,肠杆菌、肠球菌和产气荚膜梭菌均减少。结论食消饮能通过调控小肠C-kit表达、胃肠动力相关激素分泌、肠道菌群丰度、肠道黏膜结构等改善食积模型小鼠的胃肠动力与功能。Objective To determine the pharmacological effect of Shixiao decoction on mice with food retention.Methods This project consisted of experiment A and B.Totally 120 ICR mice were randomly divided into experiment A and B.Both experiments included a normal group,a food retention model group,Shixiao decoction groups(low-,medium-and high-dose),and a Jianwei Xiaoshi tablet group.After establishing the food retention model,we gave the mice corresponding test drugs by gavage twice daily for 7 days.The organ index,serum biochemical indicators,intestinal immunohistochemistry,colonic HE staining,and intestinal flora were detected in experiment A.Gastric emptying rate and intestinal propulsion rate were evaluated in experiment B.Results Compared with the model group,the spleinc organ index in the high-dose group was increased,the serum biochemical indicators in the medium-and high-dose groups were much improved.Their intestinal propulsion rate was also increased,while gastric residual rate was decreased.The immunohistochemical test showed that the expression of C-kit protein in the small intestine was much reduced in the model group,while that was increased in Shixiao decoction medium-and high-dose group and Jianwei Xiaoshi tablet group.Moreover,HE displayed that the length of colonic villi was increased,the depth of colonic crypt decreased,and the ratio of colonic villi length to crypt depth increased in both the medium-and high-dose groups.Lactobacillus and Bifidobacterium were increased,while Enterobacteriaceae,Enterococcus,and Clostridium perfringens were decreased in the high-dose group based on the fecal microbiota count.Conclusion Shixiao decoction can improve the gastrointestinal motility and function in mice with food retention mice by regulating the expression of intestinal C-kit protein,secretion of gastrointestinal motility related hormones,abundance of intestinal flora,the structure of intestinal mucosa.
