摘 要:目的建立鸭跖草药材、饮片、标准汤剂与配方颗粒的超高效液相色谱法(UPLC)特征图谱,并测定香草酸、荭草苷、异荭草苷、牡荆苷、芦丁成分的含量,考察这5个化学成分的传递与差异。方法采用Waters ACQUITY UPLC HSS T3(1.8μm,100 mm×2.1 mm)色谱柱,以甲醇和0.1%甲酸溶液为流动相梯度洗脱,流速为0.3 mL·min^(-1),柱温为30℃。以含量、特征图谱共有峰传递数及相似度为主要评价指标,分析其量值传递规律。结果15批鸭跖草药材、饮片、标准汤剂及3批配方颗粒特征图谱均呈现9个共有特征峰,与各自对照特征图谱相似度均大于0.90,指认出香草酸、荭草苷、异荭草苷、牡荆苷、芦丁5个成分,并测定其含量,未出现离散数据。结论鸭跖草药材、饮片、标准汤剂与配方颗粒的主要化学成分基本相同,UPLC特征图谱相关性良好,5个成分在测定范围内线性关系良好。所建立质量评价模式能够反映鸭跖草4种形态多成分的整体面貌,为鸭跖草配方颗粒质量控制提供数据支撑。Objective To establish an ultra performance liquid chromatography(UPLC)for the characteristics of Commelina communis and its decoction pieces,standard decoction and formula granules,and to determine the content of vanillic acid,orientin,isoorientin,vitexin,and rutin,and the transfer and difference of the five chemical components.Methods The chromatography was performed on a Waters ACQUITY UPLC HSS T3(1.8μm,100 mm×2.1 mm)with methanol(A)and 0.1% formic acid solution(B)as the mobile phase in gradient elution.The flow rate was 0.3 mL·min^(-1),and the column temperature was 30℃.The content,common peak transfer number and similarity of characteristic map were used as the main evaluation indexes.The law of magnitude transfer was determined.Results Nine common characteristic peaks were detected in the characteristic maps of 15 batches of Commelina communis and its decoction pieces,standard decoction and 3 batches of formula granules,with similarity between them and the control characteristic maps greater than 0.90.Five components(vanillic acid,orientin,isoorientin,vitexin,and rutin)were identified and their content determined without discrete data.Conclusion The main chemical components of Commelina communis and its decoction pieces,standard decoction and formula granules are basically the same,with good correlation of UPLC characteristic map.The linear relationship of the above 5 components is good in the determination range.The established evaluation model can reflect the overall quality of the four forms and multi-components of Commelina communis,providing data support for the quality control of formula granules of Commelina communis.
