摘 要:目的探讨五味珍菊片对四氯化碳(CCl_(4))诱导和急性酒精诱导小鼠肝损伤的保护作用。方法小鼠称重后分为空白组(蒸馏水)、模型组(蒸馏水)和五味珍菊片给药组(1.0、2.0、4.0 g·kg^(-1)),连续给药30 d后,模型组和五味珍菊片给药组给予5 mL·kg^(-1)的1%CCl_(4)诱发急性肝损伤模型。生化试剂盒测定小鼠血清中谷丙转氨酶和谷草转氨酶活力;HE染色法评价肝脏病理组织学情况。另将50只雄性小鼠称重后随机分成空白组(蒸馏水)、模型组(蒸馏水)、五味珍菊片给药组(1.0、2.0、4.0 g·kg^(-1)),于第30日将模型组及五味珍菊片组以12 mL·kg^(-1)剂量一次灌胃给予50%乙醇,建立急性酒精肝损伤小鼠模型。取肝组织,检测肝组织丙二醛、还原型谷胱甘肽、三酰甘油等指标,并进行病理组织学检查。结果与模型组比较,各剂量五味珍菊片均能显著降低CCl_(4)诱导肝损伤模型小鼠的血清谷丙转氨酶水平(P<0.05)、减轻肝组织损伤程度;能明显降低酒精诱导肝损伤小鼠的肝组织丙二醛、三酰甘油等含量,减轻肝组织的损伤程度。结论五味珍菊片对CCl_(4)和酒精致小鼠急性肝损伤均有辅助保护作用。Objective To determine the protective effect of Wuwei Zhenju tablets(WWZJT)on CCl_(4)-induced and acute alcohol-induced liver injury in mice.Methods The mice were divided into a blank group(distilled water),a model group(distilled water)and 3 WWZJT groups(1.0,2.0,and 4.0 g·kg^(-1))after the weighing.After 30 days of administration,the mice in the model group and WWZJT groups were given 1%CCl_(4)(5 mL·kg^(-1))to induce the acute liver injury model.The contents of alanine aminotransferase and aspartate aminotransferase in the serum was determined by biochemical kit.The histopathological changes in the liver were evaluated by HE staining.In addition,50 male mice were randomly divided into a blank group(distilled water),a model group(distilled water)and 3 WWZJT groups(1.0,2.0,and 4.0 g·kg^(-1)).On the 30th day,the model group and WWZJT groups were given 50%ethanol at 12 mL·kg^(-1) to establish a mouse model of acute alcoholic liver injury.The level of malondialdehyde,reduced glutathione,triglyceride in the liver tissues of mice in each group were detected,and the histopathology were examined.Results Compared with the model group,WWZJT significantly reduced the level of alanine aminotransferase(P<0.05)and the degree of liver tissue injury in mice with CCl_(4) induced liver injury.It also reduced the content of malondialdehyde and triglyceride in the liver tissue of mice with alcohol-induced liver injury and the degree of liver tissue damage.Conclusion WWZJT has an auxiliary protective effect on both CCl_(4)-induced and alcohol-induced liver injury.
