维生素D对糖尿病前期小鼠认知功能障碍的影响

摘  要:目的:探讨维生素D对高脂饮食诱导的糖尿病前期小鼠认知功能的影响。方法:10只4周龄的雄性C57BL/6J小鼠作为空白对照(SC)组,给予普通饲料,40只KKAy小鼠给予高脂饲料喂养6周建立糖尿病前期模型,造模成功后,糖尿病前期小鼠随机分为模型对照(MSC)组和维生素D低、中、高剂量干预组。16周后,对各组小鼠进行Morris水迷宫实验和Barnes迷宫实验;检测各组小鼠糖脂代谢相关指标;HE和尼氏染色法观察脑组织病理学变化;免疫组化法观察海马组织中AβOs和p-Tau蛋白表达变化。结果:与SC组相比,MSC组小鼠脂代谢紊乱,脑指数、空间学习和记忆能力下降,海马中的神经元数量减少、神经细胞结构紊乱,海马组织中AβOs和p-Tau蛋白表达增加(P<0.05)。与MSC组小鼠相比,各干预组小鼠认知能力、神经元数量和结构等指标有所恢复,AβOs和p-Tau蛋白表达降低(P<0.05)。结论:维生素D对高脂饮食诱导的糖尿病前期小鼠认知功能有保护作用。Aim:To explore the effects of vitamin D on cognitive function in prediabetes mice induced by high-fat diet.Method:Ten 4-week-old male C57BL/6J mice were designated as the solvent control(SC)group and provided with standard diet,while 40 KKAy mice were fed with high-fat diet for 6 weeks to establish prediabetes model.After successful modeling,prediabetic mice were randomly allocated into model solvent control(MSC)group,low,medium and high dose vitamin D intervention groups.After 16 weeks,Morris water maze test and Barnes maze test were conducted in each group.Serum glucose and lipid metabolism-related indicators were detected,pathological changes in brain tissue were observed by HE and Nissl staining,and immunohistochemical staining was used to observe changes of AβOs and p-Tau protein in hippocampal tissue.Results:Compared with the SC group,mice in the MSC group had disordered lipid metabolism,decreased brain index,spatial learning,and memory abilities,decreased number of neurons in the hippocampus,and disordered nerve cell structure,while AβOs and p-Tau protein levels in the hippocampal region were significantly increased(P<0.05).Compared with the MSC mice,the cognitive ability,number and structure of neurons in each intervention group were somewhat restored,along with significant reductions in AβOs and p-Tau protein(P<0.05).Conclusion:Vitamin D exhibits protective effects against cognitive impairment in prediabetic mice induced by high-fat diet.
