摘  要:生长调节因子(Growth-regulating factor,GRF)是一类植物特有的转录因子,在植物生长发育、激素信号转导、逆境胁迫响应等多个生物学进程中均发挥重要作用.前期研究在对甜橙GRF家族进行全基因组挖掘与逆境胁迫下表达模式分析的基础上,筛选出一个在多重激素和非生物胁迫处理下均显著诱导上调的关键基因CsGRF04.为了进一步验证CsGRF04的转录激活活性,利用酵母系统与双荧光素酶活性分析(LUC)实验分别进行分析.结果表明,CsGRF04在酵母系统中无法激活报告基因表达从而使酵母细胞在缺陷培养基中正常生长,但在LUC系统中能够显著抑制双荧光素酶活性,说明CsGRF04是一个典型的转录抑制因子.为后续深入研究CsGRF04基因在甜橙抗逆响应中的功能奠定了基础.Growth-regulating factor(GRF)is a class of plant-specific transcription factors(TFs)that play essential roles in various biological processes such as plant growth and development,hormone signal transduction,and adverse stress response.In a previous study,based on genome-wide identification and expression pattern analysis of the CsGRF family in sweet orange under adversity stress,a key gene,CsGRF04,which was significantly up-regulated under multiple phytohormone and abiotic stress treatments,was screened out.To further validate the transcriptional activation activity of CsGRF04,the yeast system and dual luciferase activity assay(LUC)were utilized in this study,respectively.The results showed that CsGRF04 was unable to activate the expression of report genes in the yeast system thus allowing yeast cells to grow normally in the deficient medium,and was able to significantly inhibit the dual-luciferase activity in the LUC system,suggesting that CsGRF04 is a typical transcriptional repressor.The study lay the foundation for subsequent in-depth research on the function of CsGRF04 in the stress response of sweet orange.
