摘  要:目的:探讨IL-6和IL-10及其基因多态性与新生儿感染性疾病(NID)之间的关系。方法:收集2022年10月至2023年6月在温州市人民医院诊断的50例NID患儿为病例组,20例健康对照样本为对照组。检测了IL-6、IL-10水平以及IL-6、IL-10基因区域的单核苷酸多态性(SNP),并进行Hardy-Weinberg平衡性检验、等位基因频率分析、χ2检验及相关性分析等统计学处理,以评估SNP与疾病和炎症因子表达水平的关联。结果:在病例组中,IL-6表达水平显著高于对照组(P<0.05)。相关性分析显示,rs1518111和rs1554286等位基因频率与IL-6表达呈显著正相关(r=0.377,P=0.007;r=0.399,P=0.004)。进一步的基因型分析表明,携带rs1518111CC基因型和rs1554286AA基因型的个体表现出更高的IL-6表达水平(t=2.819,P=0.007;t=3.012,P=0.004)。结论:IL-6表达升高在NID的发生中起着重要作用。IL-6基因区域内的rs1518111和rs1554286位点多态性可显著影响IL-6的表达水平,调控炎症反应过程,从而可能改变新生儿对感染的易感性和病情严重程度。Objective:To explore the relationship between IL-6 and IL-10 and their genetic polymorphisms as well as neonatal infectious diseases.Methods:Data from 50 neonates diagnosed with infectious diseases and 20 healthy controls were collected from Wenzhou People’s Hospital between October 2022 and June 2023.Levels of IL-6 and IL-10,as well as SNP genotyping within their gene regions,were analyzed.Statistical analysis included Hardy-Weinberg equilibrium testing,allele frequency analysis,χ2 tests,and correlation analysis to assess the association between SNPs,disease occurrence,and cytokine expression levels.Results:IL-6 levels were significantly higher in the case group compared with the control group(P<0.05).Correlation analysis revealed a significant positive correlation between allele frequencies of rs1518111 and rs1554286 and IL-6 expression levels(r=0.377,P=0.007;r=0.399,P=0.004).Further genotype analysis indicated that individuals carrying the rs1518111 CC genotype and rs1554286 AA genotype exhibited higher levels of IL-6 expression(t=2.819,P=0.007;t=3.012,P=0.004).Conclusion:Elevated expressions of IL-6 and IL-10 play significant roles in the development of neonatal infectious diseases.Polymorphisms at rs1518111 and rs1554286 within the IL-6 gene region significantly influence IL-6 expression,modulate the inflammatory response,which may alter susceptibility to infections and severity of the condition in neonates.
