摘  要:绿原酸(chlorogenic acid, CGA)是金银花(Lonicera japonica Thunb.)重要的功效成分,具有抗菌、抗病毒、防癌、防衰老等作用。本研究参照17个物种中33个绿原酸基因编码蛋白的氨基酸序列,通过NCBIBLAST扫描金银花蛋白组数据库,结合亲缘关系分析和结构域分析,筛选获得5个调控金银花绿原酸候选基因,均为Phenylalanine amononia-lyase,分别命名为LjPAL1、LjPAL2、LjPAL3、LjPAL4、LjPAL5。理化性质分析和亚细胞定位发现,这5个候选基因编码的氨基酸个数为686~714,理论等电点为5.85~6.26,均为稳定的亲水性蛋白,均定位在细胞质中。本研究为金银花的分子标记辅助育种提供基因资源,为金银花绿原酸合成代谢途径的研究提供科学依据。C hlorogenic acid is an import ant functional component of Lonicera japonica Thunb., which remain effects of antibacterial, antiretroviral, anti-cancer and anti-aging. In this study, we identified five candidate genes of Phenylalanine amononia-lyase regulating chlorogenic acid in Lonicera japonica Thunb., which were respectively named LjPAL1, LjPAL2, LjPAL3, LjPAL4, and LjPAL5, by scanning proteome database of Lonicera japonica Thunb. based on the amino acid sequences of 33 chlorogenic acid proteins from 17 species, through NCBI-BLAST,phylogenetic analysis and domain analysis. The subcellular localization, physical properties and chemical analysis showed that the five candidate genes encoded proteins with 686~714 amino acids, theoretical isoelectric points were 5.85~6.26, and they were stable hydrophilic protein localized in the cytoplasm. This study provides genetic resources for molecular marker-assisted breeding of Lonicera japonica Thunb., and provides a basis for pathways research of chlorogenic acid in Lonicera japonica Thunb..
