摘  要:本试验旨在探究维生素和益生菌复合添加剂对种公鸡精液品质及睾丸机能重塑的影响。选取50只体重[(1.49±0.24)kg]相近、健康的350日龄贵妃种公鸡,禁食15 d后随机分为2组,每组25只。对照组在恢复期饲喂基础饲粮,试验组在恢复期饲喂基础饲粮+200 mg/kg维生素和益生菌复合添加剂。试验分为预试期7 d、禁食期15 d和恢复期32 d,共计54 d。记录2组禁食前1天(F0)、禁食第15天(F15)和恢复饲喂第32天(R32)时公鸡的体重以及R32时的睾丸重、精子密度和精子活力,并对R32时的睾丸组织进行高通量转录组测序(RNA-seq)。结果显示:1)在R32时,试验组公鸡体重高于对照组(P>0.05);试验组公鸡精子活力显著高于对照组(P<0.05),睾丸重和精子密度差异不显著(P>0.05)。2)对2组公鸡睾丸组织进行RNA-seq分析,共获得57个差异基因,其中9个基因上调,48个基因下调,差异基因主要被注释到跨膜受体蛋白酪氨酸激酶激活剂活性、JAK信号转导接头活性等功能以及精囊形态发生和肌动蛋白丝束组件等过程;主要富集在甲型流感、碱基切除修复、MAPK信号通路和产生免疫球蛋白A的肠道免疫网络通路中。综上可知,维生素和益生菌复合添加剂可以通过加快种公鸡体重恢复、提高精子活力改善精液品质;通过对睾丸组织进行RNA-seq分析,筛选出T细胞急性淋巴细胞白血病1(TAL1)、类固醇激素合成急性调节蛋白(STAR)、晚疫病抗性基因(BLB1)、血管生成素4(ANGPT4)、Nei核酸内切酶Ⅷ样蛋白3(NEIL3)等基因可作为研究种公鸡精液品质和睾丸机能的候选基因。This experiment was conducted to investigate the effects of vitamin and probiotic composite additive on the semen quality and testicular function remodeling of breeding roosters.Fifty healthy 350-day-old female Guifei breeding roosters with similar body weight[(1.49±0.24)kg]were selected and randomly divided into two groups after fasting for 15 days,and each group had 25 roosters.Roosters in control group were fed a basal diet during the recovery period,and roosters in experimental group were fed the basal diet 200 mg/kg vitamin and probiotic composite additive during the recovery period.The experiment was divided into pre-feeding period for 7 days,fasting period for 15 days and recovery period for 32 days,for a total of 54 days.The body weight of the two groups at the day before fasting(F0),15 days after fasting(F15)and 32 days after resuming feeding(R32),as well as testicular weight,sperm density and sperm motile at R32 were recorded,and high throughput transcriptomic sequencing(RNA-seq)was performed on the testicular tissue at R32.The results showed as follows:1)at the R32,the body weight of experimental group was significantly higher than that of control group(P>0.05);the sperm motility of experimental group was significantly higher than that of control group(P<0.05),but the testicular weight and sperm density were not significantly different between experimental group and control group(P>0.05).2)RNA-seq analysis was performed on the testis tissues of the two groups,and 57 differential genes were obtained,of which 9 genes were up-regulated and 48 genes were down-regulated.The differential genes were mainly annotated to transmembrane receptor protein tyrosine kinase activator activity,JAK pathway signal transduction adaptor activity,seminal vesicle development,actin filament bundle assembly,and so on.It is mainly concentrated in influenza A,base excision repair,MAPK signaling pathway and intestinal immune network for immunoglobulin A production pathways.In conclusion,the vitamin and probiotic composite additi
