高效液相色谱法测定翻白草不同部位芦丁和紫云英苷的含量

摘　　要：目的:建立高效测定翻白草不同部位芦丁和紫云英苷含量的方法。方法:InertSustain^(®)C_(18)(150 mm×4.6 mm,5μm)色谱柱,甲醇-0.3%磷酸溶液为流动相,梯度洗脱,进样量10μL,检测波长349 nm,流速1.0 mL/min,柱温30℃,分别对翻白草的全草、叶和根进行芦丁和紫云英苷含量测定。结果:芦丁进样量在83.8~753.8μg时与峰面积呈良好的线性关系(R^(2)=0.9997),平均回收率为97.57%;紫云英苷进样量在82.9~746.2μg时与峰面积呈良好的线性关系(R^(2)=0.9992),平均回收率为98.16%。翻白草的全草、叶和根中芦丁含量分别为0.7105 mg/g、0.4779 mg/g、0.0717 mg/g;紫云英苷含量分别为0.6213 mg/g、0.2837 mg/g、0.0653 mg/g。结论:翻白草芦丁和紫云英苷主要集中在叶片部位;所建立的方法简便、快速、分离效果好、稳定性好,适用于翻白草中芦丁、紫云英苷的含量测定。Objective:To establish an efficient method for determine the content of rutin and astragaline in different parts of Potentilla discolor Bge..Methods:Inertsustain^(®)C_(18)(150 mm×4.6 mm,5μm)column,methanol-0.3%phosphoric acid aqueous solution as mobile phase,gradient elution,injection volume of 10μL,detection wavelength of 349 nm,flow rate of 1.0 mL/min,column temperature of 30℃,the contents of rutin and porphyrin in the whole grass,leaves and roots of Potentilla discolor Bge.are determined.Results:The linear relationship is good in the range of 83.8 to 753.8μg of rutin(R^(2)=0.9997),and the average recovery rate is 97.57%;the linear relationship is good in the range of 82.9 to 746.2μg of astragaline(R^(2)=0.9992),and the average recovery rate is 98.16%.The contents of rutin in whole grass,leaves and roots are 0.7105 mg/g,0.4779 mg/g and 0.0717 mg/g,respectively;the contents of astragaline are 0.6213 mg/g,0.2837 mg/g and 0.0653 mg/g.Conclusion:The rutin and astragaline are mainly concentrated in the leaf parts;the established method is simple,rapid,with good separation effect and stability,and is suitable for the determination of rutin and astragaline content in the Potentilla discolor Bge..
