摘 要:运动发酵单胞菌(Zymomonas mobilis)是一种唯一通过ED途径兼性厌氧发酵葡萄糖的微生物食品安全级菌株(GRAS),具有许多独特的生理特征与优良的工业生产特性.基于合成生物学方法和代谢工程改造,综述运动发酵单胞菌独特的生理特性及其在食品、医药等不同领域的应用,重点介绍运动发酵单胞菌益生效应因子及其涉及的拮抗效应、群体感应等方面的研究进展及瓶颈.同时探讨持续开发、完善高效精准的基因编辑技术、理性和非理性设计代谢途径定向调控方法及高通量筛选手段,在运动发酵单胞菌益生特性的发现以及生物治疗与固氮等方面的突破,推动合成生物学理论研究和实践生产的发展.Zymomonas mobilis,as a food-safe strain generally regarded as safe(GRAS),uniquely ferments glucose via Entner-Doudoroff(ED)pathway under anaerobic conditions.It possesses many unique physiological characteristics and excellent industrial production properties.Based on synthetic biology methods and metabolic engineering,this paper reviewed the unique physiological characteristics of Zymomonas mobilis and its applications in different fields such as food and medicine.It focused on the research progress and bottlenecks of probiotic effectors in Zymomonas mobilis,including their involvement in antagonistic effects and quorum sensing.Additionally,it also discussed the continuous development and improvement of high-efficiency and precise gene editing technologies,rational and irrational design methods for directional regulation of the metabolic pathway and high-throughput screening methods.These advancements aim to promote breakthroughs in the probiotic characteristics of Zymomonas mobilis,as well as in its application areas such as biomedicine and nitrogen fixation,thereby driving the development for theory research and practical production of the synthetic biology.
