摘 要:目的通过90d经口毒性试验观察浙产山茱萸对SD大鼠的健康影响。方法SD大鼠随机分成4组,包括1个阴性对照组和低、中、高3个剂量组。阴性对照组喂饲基础饲料,3个剂量组按2.0、4.0和8.0g/kg·bw设计,分别在饲料中掺入粉碎的山茱萸,连续喂饲13周。观察动物间体重、进食量等一般状况的变化,在试验中期和末期检测血液学和血清生化指标及尿常规,试验期满测定脏器重量,计算脏体比,并对主要脏器进行病理组织学观察。结果试验期内3个剂量组雌鼠和低、中剂量组雄鼠体重和进食情况与阴性对照组比较,差异均无统计学意义(P>0.05)。经Bonferroni法两两比较,高剂量组雄性大鼠前半程体重较阴性对照组有明显下降(F=20.657,P_(校正)=0.004),经Tukey法两两比较,高剂量组雄性大鼠总食物利用率较阴性对照组有明显下降(Q=3.767,P=0.001)。各组大鼠血液学、血清生化、尿常规指标均无明显异常,主要脏器重和脏体比与阴性对照组比较差异无统计学意义(P>0.05),主要脏器均未发现明显的病理组织学改变。结论浙产山茱萸大鼠亚慢性经口毒性试验NOAEL值为8.0g/kg·bw,但该剂量可能影响雄性大鼠的体重增长,其生物学意义有待进一步研究。ObjectiveTo learn the influence of Corni Fructus originated from Zhejiang on the health of SD rats by the 90-day oral toxicity test.Methods SD rats were randomly divided into 4 groups including a control group and three experimental groups.The rats of the control group were fed on the basal feed.The rats of the experimental groups were fed on the feed mixed by Corni Fructus with different mass percentages.The designed dose of the experimental groups were 2.O,4.0 and 8.0 g/kg·bw.Body weight and food consumption of each groups were observed.The haematological and serum biochemical parameters and urinalysis were measured at the middle and the end of the test.The main organ tissue analysis including organ weight,the ratio of organ and body weight and the histopathological examination were also performed.Results There were no significant differences of body weight and food intake in female and male rats of three experimental groups except the high dose group of male rats compared to the control groups.Body weight throughout the first period in high dose group of male rats was decreased compared to the control group by pairwise comparison using Bonferroni method(F=20.657,P_(校正)=0.004).Total food consumption in high dose group of male rats was reduced compared to the control group by pairwise comparison using Tukey's honestly significant dfference test(Q=3.767,P=0.001).No significant differences of the haematological,serum biochemical and urinal indices,analysis of the main organ tissue were shown in rats of three experimental groups compared to control group.Conclusion The NOAEL of sub-chronic oral toxicity test of Corni Fructus originated from Zhejiang in rats is 8.O g/kg·bw.But the biological significance of the effect of this dose on weight gain of male rats needs further research.
