摘  要:目的研究Ghrelin通过ERK1/2信号通路促进大鼠骨髓间充质干细胞向软骨分化的机制。方法骨髓间充质干细胞(BMSCs)在适当的刺激下可分化为软骨母细胞、脂肪细胞或软骨细胞。生长激素释放肽(Ghrelin)是生长激素促分泌肽受体的内源性配体,能够刺激生长激素的分泌。为研究能够触发BMSCs向软骨高效分化机制,寻找治疗异位骨化的新型治疗方案。本文采用600 ng/mLGhrelin培养r BMSCs,通过免疫蛋白印记法检测磷酸化状态ERK1/2、p38、JNK以及Ⅱ型胶原的变化。结果使用600 ng/mL Ghrelin作用于BMSCs 24 h,Ⅱ型胶原的表达达到最高水平;Ghrelin主要通过ERK1/2信号通路诱导r BMSCs向软骨分化。结论本研究可为揭示ERK1/2信号通路在Ghrelin诱导的r BMSCs生长中的潜力提供医学参考意义。Objective To investigate the mechanism of Ghrelin promoting the differentiation of rat bone marrow mesenchymal stem cells into cartilage through ERK1/2 signaling pathway.Methods Bone marrow mesenchyml stem cell(BMSCs)can differentiate into chondroblasts,adipocytes or chondrocytes under appropriate stimulation.Ghrelin is an endogenous ligand of growth hormone secretagogue peptide receptor,which can stimulate the secretion of growth hormone.In order to study the mechanism that can trigger the efficient differentiation of bone marrow mesenchyml stem cell into cartilage,and improve the development of novel treatment for ectopic ossification.rBMSCs were cultured with 600 ng/mLGhrelin,and the phosphorylation of ERK1/2,p38,JNK and type II collagen were detected by immunoprotein imprinting.Results The expression of type II collagen reached the highest level when 600 ng/mL growth hormone releasing peptide was applied to rBMSCs for 24 h.Ghrelin induces rat bone marrow mesenchyml stem cell to differentiate into cartilage mainly through ERK1/2 signaling pathway.Conclusions This study can provide medical significance for revealing the potential of ERK1/2 signaling pathway in the growth of rBMSCs induced by Ghrelin.
