摘  要:小泛素相关修饰物(SUMO)作为泛素类似物家族的一员对多种底物蛋白修饰发挥生物学作用,其以酶为媒介与靶点蛋白结合产生生化级联反应,与细胞增殖凋亡、转录复制及炎症信号通路调控密切相关。类风湿关节炎(RA)是以关节滑膜炎症为主要病理表现的慢性自身免疫性疾病。目前,国内外对SUMO化修饰在RA抗炎机制中的研究较少,其机制可能是通过参与核因子κB信号通路、转化生长因子-β信号通路及促分裂原活化的蛋白激酶信号通路中重要炎症因子的激活和释放,修饰多条RA相关炎症信号通路中的关键蛋白底物,从微观上保持免疫稳态平衡,以抑制炎症的发生发展。未来,深入研究SUMO化修饰在RA发病机制中的作用,可为RA靶向治疗药物的开发提供参考和借鉴。As a member of the ubiquitin analog family,small ubiquitin-related modifiers(SUMO)play biological roles in a variety of substrate protein modifications.They combine with target proteins to produce biochemical cascade,which is closely related to cell proliferation,apoptosis,transcription replication and regulation of inflammatory signaling pathways.Rheumatoid arthritis(RA)is a chronic autoimmune disease with synovial inflammation of the joints as the main pathological manifestation.At present,the domestic and foreign studies on SUMO modification in RA anti-inflammatory mechanism are few,which may be by participating in the activation and release of important inflammatory factors in nuclear factor-κB signaling pathway,transforming growth factor-βsignaling pathway and mitogen-activated protein kinase signaling pathway,modifying key protein substrates in multiple in RA related inflammatory signaling pathways,maintaining microscopic immune balance to inhibit the occurrence and development of inflammation.In the futre,in-depth research the role of SUMO modification in RA pathogenesis can provide references for the development of targeted therapeutic drugs for RA.
