摘  要:新建一种采用HPLC–RID,同时测定银杏叶中奎宁酸、莽草酸含量的方法,评价不同树龄和采摘期对银杏叶中2种物质含量的影响。结果表明:银杏叶中奎宁酸和莽草酸分别在0~250.00μg/mL和0~121.76μg/mL内呈良好的线性关系,回归方程分别为:Y=108.83X-182.19(r=0.9930)和Y=132.125X+0.19(r=0.9992),平均回收率分别为99.47%和99.40%,RSD分别为0.94%和1.32%。三年生、四年生银杏叶中奎宁酸和莽草酸含量较其他树龄的高,分别达77.39~58.57 mg/g和76.80~52.37 mg/g;4—5月采摘的银杏叶中奎宁酸和莽草酸含量较其他采摘月份高,分别达70.35~72.91 mg/g和71.06~72.44 mg/g。综合可知新方法重复性好、操作简便、快速稳定、易于普及,同时能有效测定银杏叶中奎宁酸和莽草酸含量,且探明了奎宁酸和莽草酸的工业生产最优原料为三年生、四年生及4—5月采摘的银杏叶。A HPLC-RID method was established to determine the content of quinic acid and shikimic acid in Ginkgo biloba leaves,and the effects of different tree ages and harvesting months on their content were evaluated.The results showed that quinic acid and shikimic acid had good linear relationships within the ranges of 0-250.00μg/mL and 0-121.76μg/mL,with the regression equations of Y=108.83X-182.19(r=0.9930)and Y=132.125X+0.19(r=0.9992),average recovery of 99.47%and 99.40%,and RSD of 0.94%and 1.32%,respectively.Moreover,the content of quinic acid and shikimic acid in the leaves of 3 to 4-year-old G.biloba was higher than that in the plants of other years old,reaching 77.39-58.57 mg/g and 76.80-52.37 mg/g,respectively.In addition,the content of quinic acid and shikimic acid in G.biloba leaves harvested from April to May was higher than that harvested in other months,reaching 70.35-72.91 mg/g and 71.06-72.44 mg/g,respectively.A method for simultaneous determination of quinic acid and shikimic acid in G.biloba leaves was established in this study,with good repeatability,simple operation,short determination time,reliable results,and easy popularization.Furthermore,this study proved that the optimal raw materials for industrial production of quinic acid and shikimic acid were leaves from G.biloba of 3 to 4 years old and harvested from April to May.
