人参皂苷Rh2、CK对卵巢癌细胞恶性行为及对卵巢癌细胞-巨噬细胞串扰影响的研究

摘　　要：目的:探讨人参皂苷Rh2、CK对卵巢癌细胞增殖、迁移及TNFAIP8表达的影响以及对卵巢癌细胞-巨噬细胞串扰的影响。方法:培养卵巢癌OVCAR3细胞株,将其分为空白组、Rh2-Xμmol/L(30、60、100μmol/L)组、CK-Xμmol/L(20、40、60μmol/L)组,通过CCK-8法检测细胞的增殖能力,划痕实验检测细胞的迁移能力,RT-PCR法检测TNFAIP8 mRNA表达水平,Western blot法检测TNFAIP8蛋白表达水平。获得肿瘤相关巨噬细胞(TAM),将其分为OVCAR3组、TAM组、OVCAR3+TAM组、OVCAR3+TAM+Rh2-100μmol/L组、OVCAR3+TAM+CK-60μmol/L组,通过CCK-8法检测OVCAR3细胞的增殖能力,划痕实验检测OVCAR3细胞的迁移能力,流式细胞术分别检测TAM细胞CD206和CD163的阳性细胞数。结果:人参皂苷Rh2、CK处理后,显著抑制了OVCAR3细胞的增殖和迁移活性(P<0.05),并且呈现出浓度依赖性,TNFAIP8 mRNA及蛋白表达水平显著下降(P<0.05);OVCAR3+TAM组较OVCAR3组OVCAR3细胞增殖和迁移能力显著增加(P<0.05),较TAM组CD206和CD163阳性的TAM细胞数显著增加,人参皂苷Rh2、CK处理后,抑制了OVCAR3+TAM组中OVCAR3细胞增殖和迁移(P<0.05),CD206和CD163阳性的TAM细胞数显著下降(P<0.05)。结论:人参皂苷Rh2、CK对卵巢癌细胞恶性行为及对卵巢癌细胞-巨噬细胞串扰有抑制作用。Objective:To explore the effects of Ginsenosides Rh2 and CK on the proliferation,migration,and TNFAIP8 expression of ovarian cancer cells,as well as their impact on the crosstalk between ovarian cancer cells and macrophages.Methods:Ovarian cancer OVCAR3 cell lines were cultured and divided into control group,Rh2-Xμmol/L(30,60,100μmol/L)groups,and CK-Xμmol/L(20,40,60μmol/L)groups.Cell proliferation was assessed using the CCK-8 assay,cell migration was evaluated through wound healing assays,and TNFAIP8 mRNA and protein levels were measured by RT-PCR and Western blot,respectively.Tumor-associated macrophages(TAMs)were obtained and divided into the OVCAR3 group,TAM group,OVCAR3+TAM group,OVCAR3+TAM+Rh2-100μmol/L group,and OVCAR3+TAM+CK-60μmol/L group.The proliferation and migration of OVCAR3 cells and the positive cell counts for CD206 and CD163 in TAMs were analyzed using the CCK-8 assay,wound healing assays,and flow cytometry.Results:Treatment with Ginsenosides Rh2 and CK significantly inhibited the proliferation and migration of OVCAR3 cells(P<0.05)in a concentration-dependent manner,with a concomitant decrease in TNFAIP8 levels(P<0.05).In the OVCAR3+TAM group,there was a significant increase in the proliferation and migration of OVCAR3 cells compared to the OVCAR3 group(P<0.05),and an increase in CD206 and CD163 positive TAM cells compared to the TAM group.Treatment with Ginsenosides Rh2 and CK inhibited the proliferation and migration in the OVCAR3+TAM group(P<0.05)and decreased the counts of CD206 and CD163 positive TAM cells significantly(P<0.05).Conclusion:Ginsenosides Rh2 and CK inhibit the malignant behaviors of ovarian cancer cells and the crosstalk between ovarian cancer cells and macrophages.
