三七总皂苷调控miR-22对高血脂合并冠心病大鼠脂质代谢和氧化应激的机制研究

摘  要:本研究旨在探讨三七总皂苷能否通过miR-22调节高血脂合并冠心病大鼠脂质代谢和氧化应激水平。试验将60只大鼠随机分为6组,即Blank组、HLP+CHD组、PNS组、miR-NC组、miR-22组和PNS+anti-miR-22组,10只/组。检测大鼠心功能;RT-PCR检测心肌组织中miR-22相对表达量;检测血脂指标和氧化应激指标;HE染色检测心肌组织病理学变化;蛋白质印迹法检测心肌组织中Nrf2、HO-1蛋白表达。结果表明,HLP+CHD组和miR-NC组心肌组织中miR-22相对表达量、LVEF、血清HDL-C水平、SOD、GSH-Px、CAT水平及心肌组织中Nrf2、HO-1蛋白表达均低于Blank组,LVESD、LVEDD、血清TC、TG、LDL-C水平、MDA水平均高于Blank组(P<0.05);和HLP+CHD组相比,PNS组和miR-22组心肌组织中miR-22相对表达量、LVEF、血清HDL-C水平、SOD、GSH-Px、CAT水平及心肌组织中Nrf2、HO-1蛋白表达明显升高,LVESD、LVEDD、血清TC、TG、LDL-C水平、MDA水平明显降低(P<0.05);PNS+anti-miR-22组心肌组织中miR-22相对表达量、LVEF、血清HDL-C水平、SOD、GSH-Px、CAT水平及心肌组织中Nrf2、HO-1蛋白表达均低于PNS组,LVESD、LVEDD、血清TC、TG、LDL-C水平、MDA水平均高于PNS组(P<0.05)。三七总皂苷可通过调节miR-22水平,改善高血脂合并冠心病大鼠脂质代谢紊乱和氧化应激状态。The aimof this study was to investigate whether total saponins of Panax notoginseng can regulate miR-22 to regulate lipid metabolism and oxidative stress in rats with hyperlipidemia and coronary heart disease.60 rats were randomly divided into 6 groups:Blank group,HLP+CHDgroup,PNS group,miR-NC group,miR-22 group and PNS+anti-miR-22 group(10 rats/group).The heart function of rats was detected.The relative expression of miR-22 in myocardial tissue was detected by RT-PCR.Blood lipid index and oxidative stress index were detected.Myocardial histopathological changes were detected by HEstaining.The expression of Nrf2 and HO-1 proteins in cardiac tissue was detected by western blot.The results showthat the relative expression of miR-22,LVEF,serumHDL-Clevel,SOD,GSH-Px,CAT level and Nrf2 and HO-1 protein expression in cardiac tissue of HLP+CHDgroup and miR-NCgroup were lower than those of Blank group.LVESD,LVEDD,serumTC,TG,LDL-C and MDAlevels were higher than those in Blank group(P<0.05).Compared with HLP+CHDgroup,the relative expression level of miR-22,LVEF,serumHDL-C level,SOD,GSH-Px,CAT level and Nrf2 and HO-1 protein expression in myocardial tissue of PNS group andmiR-22 group were significantly increased.The levels of LVESD,LVEDD,serumTC,TG,LDL-CandMDAwere significantly decreased(P<0.05).The relative expression level of miR-22,LVEF,serumHDL-Clevel,SOD,GSH-Px,CATlevel andNrf2 and HO-1 protein expression in myocardial tissue of PNS+anti-miR-22 group were lower than those of PNS group.The levels of LVESD,LVEDD,serum TC,TG,LDL-C and MDA were higher than those of PNS group(P<0.05).Panax notoginseng saponins can improve lipid metabolism disorder and oxidative stress in hyperlipidemia rats with coronary heart disease by regulating the level of miR-22.
