摘 要:为了探究维生素C对藏鸡精液低温及冷冻保存的作用,试验采集健康的25周龄藏鸡精液,在其稀释液或冻存液中添加不同浓度的维生素C后进行低温(4℃)与冷冻保存,检测保存后精子的活率、总超氧化物歧化酶(total superoxide dismutase,T-SOD)活力、丙二醛(malondialdehyde,MDA)含量。结果表明:用添加0.01 mg/mL维生素C的稀释液在4℃保存24 h和48 h后,藏鸡精子的活率显著提高(P<0.05),精子T-SOD活力有升高的趋势,而MDA含量有下降的趋势;冻存液中添加0.01 mg/mL的维生素C,冷冻解冻后精子活率及线粒体活性显著提高(P<0.05),同时精子T-SOD活力显著提高(P<0.05),MDA含量显著降低(P<0.05)。说明低温或冷冻保存藏鸡精液时添加0.01 mg/mL的维生素C可有效提高保存精子的活率及抗氧化能力。In order to explore the role of vitamin C in cold and frozen preservation of semen in Tibetan chicken,in the experiment,the semen of healthy 25-week-old Tibetan chickens was collected;different concentrations of vitamin C were added to its dilution or freezing solution for cold and frozen preservation.Sperm viability,total superoxide dismutase(T-SOD)activity and malondialdehyde(MDA)content were measured after preservation.The results showed that the sperm motility of Tibetan chicken was significantly increased after storage at 4℃for 24 h and 48 h with the dilution of 0.01 mg/mL vitamin C(P<0.05);sperm T-SOD motility tended to increase;however,the MDA content had a decreasing trend.0.01 mg/mL of vitamin C was added to the refrigerant solution,and sperm motility and mitochondrial activity were significantly increased after freezing and thawing(P<0.05).At the same time,the T-SOD motility of sperm was significantly increased(P<0.05);the content of MDA was significantly decreased(P<0.05).The results indicated that the addition of 0.01 mg/mL vitamin C to the semen of Tibetan chicken at cold and frozen preservation could effectively improve the motility and antioxidant capacity of preserved sperm.
