基于XIAP及细胞凋亡探讨经皮穴位电刺激对早发性卵巢功能不全大鼠的作用效果

摘  要:目的基于X连锁凋亡抑制蛋白(X-linked inhibitor of apoptosis protein,XIAP)及相关凋亡因子探讨经皮穴位电刺激(transcutaneous electrical acupoint stimulation,TEAS)调节早发性卵巢功能不全(premature ovarian insufficiency,POI)大鼠卵巢功能的可能机制。方法54只雌性SD大鼠随机分为空白组(n=15)、造模组(n=39),造模组以连续14 d雷公藤多苷灌胃的方法构建POI大鼠模型,空白组以生理盐水灌胃作对照,造模结束随机处死3只大鼠取卵巢判断造模效果。在造模成功后,造模组(n=36)随机分为模型组(n=12)、TEAS组(n=12)、西药组(n=12),进行为期4周的干预,TEAS组给予电子针疗仪刺激三阴交、关元和肾俞穴,西药组予戊酸雌二醇配液灌胃,同期空白组(n=12)与模型组予生理盐水灌胃。通过ELISA法测定血清卵泡刺激素(follicle stimulating hormone,FSH)、雌二醇(estradiol,E_(2))水平,TUNEL法测定卵巢组织细胞凋亡水平,Western blot检测B淋巴细胞瘤-2(B cell lymphoma-2,Bcl-2)、Bcl-2同源二聚体X(Bcl-2-Associated X,Bax)、Caspase-3、XIAP含量。结果造模组大鼠体质量下降、动情周期延长或紊乱、内分泌紊乱、卵巢形态异常提示造模成功。治疗结束后,TEAS组、西药组与空白组体质量、卵巢指数明显高于模型组(P<0.05),且动情周期恢复。与空白组比较,模型组大鼠血清E_(2)水平和卵巢组织Bcl-2、XIAP、Bcl-2/Bax表达降低(P<0.05或P<0.01),血清FSH水平、卵巢组织Bax、Caspase-3蛋白表达、细胞凋亡率升高(P<0.05或P<0.01)。与模型组相比,TEAS组和西药组大鼠血清E_(2)水平均显著升高,卵巢组织Bcl-2、XIAP、Bcl-2/Bax蛋白明显增高(P<0.05),血清FSH水平、卵巢组织Bax、Caspase-3蛋白、细胞凋亡率降低(P<0.05)。结论TEAS可调节POI大鼠内分泌激素水平,减少卵巢组织细胞凋亡,这种作用机制可能与上调XIAP表达有关。Objective This study explores the potential mechanism of transcutaneous acupoint electrical stimulation(TEAS)in regulating ovarian function in a rat model of premature ovarian insufficiency(POI),focusing on XIAP and related apoptotic factors.Methods Fifty-four female Sprague-Dawley(SD)rats were randomly divided into a control group(n=15)and a model group(n=39).The POI model was induced by administering tripterygium glycosides for 14 days via gavage in the model group,while the control group received normal saline.After modeling,three rats were sacrificed to assess the efficacy of the model by examining ovarian morphology.The successful model rats(n=36)were randomly divided into a model group(n=12),a TEAS group(n=12),and a Western medicine group(n=12),with interventions lasting four weeks.The TEAS group received electronic acupuncture at the Sanyinjiao,Guanyuan,and Shenshu acupoints,while the Western medicine group was given estradiol valerate solution orally.ELISA was used to measure serum follicle-stimulating hormone(FSH)and estradiol(E_(2))levels,TUNEL assays assessed ovarian tissue apoptosis,and Western blot analysis was conducted to determine the protein levels of Bax,Bcl-2,Caspase-3,and XIAP.Results POI modeling was validated by decreased body weight,prolonged or disordered estrous cycles,endocrine disruption,and abnormal ovarian morphology.Following treatment,body weight and ovarian index were significantly higher in the TEAS and Western medicine groups than in the model group(P<0.05),and the estrous cycle was restored.Compared with the control group,the model group showed decreased serum E_(2),Bcl-2,XIAP,and Bcl-2/Bax ratio(P<0.05 or P<0.01)and increased FSH,Bax,Caspase-3 protein expression,and apoptosis rate(P<0.05 or P<0.01).Compared with the model group,both the TEAS and Western medicine groups exhibited significantly elevated serum E_(2),increased Bcl-2,XIAP,and Bcl-2/Bax levels in ovarian tissues(P<0.05),and reduced FSH,Bax,Caspase-3,and apoptosis rate in ovarian tissues(P<0.05).Conclusion TEAS can re
