摘  要:目的建立薰衣草精油喷膜剂的制备工艺,并进行药效评价。方法以提取时间、乙醇体积分数、乙醇用量为考察因素,以干膏率、游离总蒽醌含量、欧前胡素含量为评价指标,采用L_(9)(3^(3))正交试验法优选大黄、白芷的提取工艺。将提取液混合后进行喷雾干燥,通过单因素试验筛选喷膜剂的成膜材料、乙醇体积分数、药粉溶解方式,制备薰衣草精油喷膜剂。对制备的薰衣草精油喷膜剂进行体外抑菌实验;构建小鼠烫伤模型,评价其药效。结果优选大黄、白芷的提取工艺为加10倍量60%乙醇提取2次,每次2.0 h,提取液混合后于120℃下进行喷雾干燥。单因素试验结果显示,成膜材料选择3%羟丙基甲基纤维素,溶剂选择80%乙醇,药粉、薰衣草精油、四针氧化锌晶须复合物直接加入溶胀的膜材中,补足溶剂,即成薰衣草精油喷膜剂。抑菌实验结果显示,薰衣草精油喷膜剂对金黄色葡萄球菌、大肠杆菌的抑菌圈为8~12 mm,抑菌作用良好。药效评价结果显示,薰衣草精油喷膜剂能加快烫伤的愈合。结论成功制备了抗菌作用良好且能促进烫伤皮肤修复的薰衣草精油喷膜剂。Objective To establish the prepare process of film spraying agent of lavender essential oil,and to evaluate its pharmaceutical.Methods With extraction time,ethanol volume fraction,and ethanol dosage as investigation factors,and with dry paste rate,free total anthraquinone content,and resveratrol content as evaluation indicators,the L_(9)(3^(3))orthogonal test was used to optimize the extraction process of Rhei Radix et Rhizoma and Angelicae Dahuricae Radix.The extract was mixed and then spray dried.The film-forming material of the spray agent,ethanol volume fraction,and the dissolving mode of the medicinal power were screened through the single factor test to prepare the film spraying agent of lavender essential oil.The in vitro antibacterial test of the prepared film spraying agent of lavender essential oil was conducted.The mouse burn model was established to evaluate its pharmacodynamics.Results The optimal extraction process of Rhei Radix et Rhizoma and Angelicae Dahuricae Radix was to extract with 10 times of 60%ethanol twice,each time for 2 h,and spray-dry the extract at 120℃after mixing.The results of the single factor test showed that 3%hydroxypropyl methyl cellulose(HPMC)was selected as film-forming material,80%ethanol was selected as solvent.The powder,lavender essential oil and four-needle zinc oxide whisker compound were directly added to the swollen film material,and then the solvent was supplemented to form the film spraying agent.The results of antibacterial experiment showed that the bacteriostatic circle of the film spraying agent of lavender essential oil on Staphylococcus aureus and Escherichia coli was 8-12 mm,with a good antibacterial effect.The results of the pharmaceutical evaluation showed that film spraying agent of lavender essential oil could accelerate the healing of burns.Conclusion Film spraying agent of lavender essential oil with a good antibacterial effect and promoting the repair of scalded skin was successfully prepared.
