益气通络方对大鼠血管内膜增生、平滑肌细胞表型转化和增殖的影响

摘　　要：目的:探讨益气通络方对大鼠颈总动脉血管内膜增生(IH)、血管平滑肌细胞(VSMCs)表型转化和增殖的影响。方法:将36只SD大鼠随机分为假手术组、模型组、益气通络方高、中、低剂量组和阿托伐他汀组,每组6只。除假手术组外,其余各组采用球囊导管术建立大鼠左侧颈总动脉内膜增生模型,各组灌胃相应药物及蒸馏水14 d。采用HE染色法观察各组大鼠颈总动脉病理变化及血管内膜增生程度;采用免疫组化法和Western blot法检测各组大鼠颈总动脉α-平滑肌肌动蛋白(α-SMA)、骨桥蛋白(OPN)和增殖细胞核抗原(PCNA)蛋白表达情况。结果:与假手术组比较,模型组颈总动脉内膜增生明显,血管内膜面积(IA)、内膜厚度(IT)、内膜面积增生率(HRIA)和内膜厚度增生率(HRIT)均显著增加(P<0.01);颈总动脉VSMCs收缩表型α-SMA表达显著降低(P<0.05),合成表型标志物OPN和细胞增殖标志物PCNA表达显著增加(P<0.01)。与模型组比较,益气通络方高剂量组及阿托伐他汀钙片组IA、IT、HRIA、HRIT均显著降低(P<0.01);阿托伐他汀组OPN表达显著降低;益气通络方中、高剂量组ɑ-SMA表达显著升高(P<0.01),PCNA表达显著降低(P<0.05)。结论:益气通络方能够有效改善SD大鼠血管内膜增生,可能与调控VSMCs表型转化及增殖有关。Objective To investigate the effects of Yiqi Tongluo decoction on common carotid artery intimal hyperplasia(IH)and phenotypic transformation and proliferation of vascular smooth muscle cells(VSMCs)in rats.Methods Thirty-six SD rats were randomly divided into sham operation group,model group,Yiqi Tongluo decoction high,medium and low dose groups and atorvastatin group,6 rats in each group.Except the sham operation group,the left common carotid artery intimal hyperplasia model was established by balloon catheters in the other groups,and the corresponding drugs and distilled water were intragastric in each group for 14 days.HE staining was used to observe the pathological changes of common carotid artery and the degree of intimal hyperplasia in each group.The expressions ofα-smooth muscle actin(α-SMA),osteopontin(OPN)and proliferating cell nuclear antigen(PCNA)in common carotid artery were detected by immunohistochemistry and Western blot.Results Compared with sham operation group,common carotid artery intimal hyperplasia was obvious in model group,and the intimal area(IA),intimal thickness(IT),intimal area hyperplasia rate(HRIA)and intimal thickness hyperplasia rate(HRIT)were significantly increased(P<0.01).The expression ofα-SMA in VSMC contraction phenotype was significantly decreased(P<0.05),and the expression of OPN and PCNA,a cell proliferation marker,was significantly increased(P<0.01).Compared with model group,IA,IT,HRIA and HRIT in Yiqi Tongluo decoction high-dose group and atorvastatin calcium tablet group were significantly decreased(P<0.01).The expression of OPN was significantly decreased in atorvastatin group.The expression of alpha SMA in the medium and high dose groups of Yiqi Tongluo decoction was significantly increased(P<0.01),while the expression of PCNA was significantly decreased(P<0.05).Conclusion Yiqi Tongluo decoction can effectively improve the hyperplasia of vascular intima and regulate the phenotypic transformation and proliferation of VSMCs in SD rats.
