基于加权基因共表达网络分析探究脑络欣通调控缺血性脑卒中炎症作用及分子机制

摘  要:目的基于加权基因共表达网络分析(weighted gene co-expression network analysis,WGCNA)探究脑络欣通治疗缺血性脑卒中(ischemic stroke,IS)的作用及分子机制。方法首先从GEO数据库获取IS临床患者转录组数据,通过limma进行差异基因分析和借助WGCNA挖掘IS相关靶点;然后筛选脑络欣通的活性成分并预测其作用靶点;最后对基于WGCNA筛选出的靶点与预测的作用靶点取交集,并取关键成分与靶点进行分子对接验证。结果WGCNA结果显示,脑络欣通活性成分作用的关键靶点有5个,即GLRX、ELANE、S100A8、BCL2A1和CAMP;KEGG富集分析显示,脑络欣通主要通过调控白细胞介素(interleukin,IL)-17和核因子(nuclear factor,NF)-κB炎症信号通路来达到抗IS的作用;分子对接结果显示,脑络欣通活性成分与其核心靶点具有良好的结合力。结论脑络欣通治疗IS具有多成分、多靶点和多通路协同作用的特征,其分子机制可能是通过脑络欣通中主要活性成分对IL-17和NF-κB炎症信号通路的调控来达到治疗IS的作用。Objective To investigate the role and molecular mechanism of Naoluoxintong Decoction in regulating inflammation in ischemic stroke(IS)using weighted gene co-expression network analysis(WGCNA).Methods GEO database was used to obtain the transcriptome data of IS patients,limma was used for the analysis of differentially expressed genes,and WGCNA was used to obtain IS-related targets.The active components of Naoluoxintong Decoction were obtained,and their action targets were predicted.The targets identified by WGCNA were intersected with the predicted action targets,and molecular docking was performed for the key components and targets.Results The WGCNA analysis identified five key targets for the active components of Naoluoxintong Decoction,i.e.,GLRX,ELANE,S100A8,BCL2A1,and CAMP.The KEGG enrichment analysis showed that Naoluoxintong Decoction mainly exerted an anti-IS effect by regulating the IL-17 and NF-κB signaling pathways.Molecular docking validated the core targets with good binding activities to the key compounds.Conclusion Naoluoxintong Decoction exerts a therapeutic effect on IS through multiple components,targets,and pathways,possibly by regulating the IL-17 and NF-κB inflammatory signaling pathways through the main active components of Naoluoxintong Decoction.
