摘 要:利用致聋方法制作白腰文鸟幼鸟聋哑鸟和成年致聋鸟模型,为研究幼鸟失聪后形成聋哑症的机制提供了一种最佳实验方法。使用血液/细胞/组织基因组DNA提取试剂盒(DP304-02)提取白腰文鸟幼鸟基因组DNA,根据电泳后约200 bp条带数确定其性别。使用细钩针勾住耳蜗的基部,将幼鸟的耳蜗拔出,并镜检拔出的耳蜗的完整性,致聋后的幼鸟会逐渐发育变成聋哑鸟。用语图软件分析鸟语的声谱与音节参数,分析幼年致聋鸟的声谱与成年致聋鸟鸣唱语句的慢慢畸变过程有利于研究聋哑形成的机制。利用电镜方法研究致聋鸟模型听觉与发声中枢的退化,为相关基础研究提供参考。The creation of deaf-mute birds through the induction of deafness in juvenile birds and the establishment of adult deafened bird models present an optimal experimental approach for investigating the mechanisms underlying deaf-mutism formation subsequent to juvenile deafness.Utilizing the Blood/Cell/Tissue Genomic DNA Extraction Kit(DP304-02)for genomic DNA extraction,the gender of juvenile birds was determined based on the number of electro⁃phoretic bands formed at 200 bp.Employing fine hooks to grasp the base of the cochlea,the cochlea of the juvenile bird was extracted,and the integrity of the extracted cochlea was examined under a microscope.Juvenile birds subject⁃ed to deafening will gradually transition into deaf-mute birds.Sonogram software was utilized to analyze the spectro⁃gram and phonetic parameters of their bird songs.Analyzing the gradual distortion process of the spectrogram of juve⁃nile deafened birds and the singing patterns of adult deafened birds was conducive to investigating the mechanism of deaf-mutism formation.Studying the degeneration of the auditory and vocal central nervous system of deafened birds using electron microscopy technology can offer valuable insights for fundamental research in this field.
