摘 要:基于阿莫西林(AMC)和克拉维酸(CLA)的吸收特性,在230 nm波长处定量AMC/CLA混合物的总浓度(c_(T)),同时在272 nm的AMC特征吸收处定量AMC浓度,通过两者之差定量CLA的浓度。基于此,开发出一种操作简便、成本低的同时测定各种相关药物中AMC和CLA的新方法。结果表明,优化的分析条件为p H 6.0的30 mmol/L磷酸盐缓冲液作为稀释液,辅料不产生干扰;AMC/CLA混合物的标准曲线为A_(T)=0.0253c_(T)+0.0214 (r=0.99999),AMC的标准曲线方程为A_(272)=0.0031c_(AMC)+0.006 (r=0.9999),AMC和CLA的相对标准偏差为0.32%~2.9%,标准添加回收率为95.3%~105.0%。本方法测定结果与高效液相色谱法结果一致,可用于制药过程中AMC/CLA混合样品的快速质量监控。Based on the absorption properties of amoxicillin (AMC) and clavulanic acid (CLA),the total concentration of mixture of AMC and CLA was quantified at 230 nm wavelength.At the same time,the AMC concentration was determined at 272 nm of the AMC characteristic absorption,and the CLA concentration was quantified by the difference between AMC/CLA and AMC concentrations.On this basis,a simple and low-cost method was developed for the simultaneous determination of AMC and CLA in various related medicines.The results showed that the optimal condition was phosphate buffer of p H 6.0 with concentration of 30 mmol/L as diluent,and the excipients has no interference on detection.The standard curve of AMC/CLA mixture was A_T=0.0253c_T+0.0214 (r=0.99999),and that of AMC was A_(272)=0.0031c_(272)+0.006 (r=0.9999).The relative standard deviations for AMC and CLA ranged from 0.32% to 2.9%,and the recoveries of standard addition experiments were 95.3%-105.0%.The results of this method were consistent with that of HPLC method,and this method can realize rapid quality monitoring for AMC/CLA mixture during production.
