摘 要:建立高效液相色谱法测定异丙醇-水溶液中利血平含量。采用Eclipse Plus C18色谱柱(150 mm×4.6 mm,5μm)分离,以甲醇-水溶液(体积比为8∶2)作为流动相等度洗脱,采用波长为268 nm的二极管阵列检测器,流量为0.5 mL/min,柱温为40℃。利血平的质量浓度在0.2~10μg/mL范围内与色谱峰面积线性关系良好,相关系数为0.9999,检出限为0.05μg/mL。利血平的加标回收率为100.20%~100.39%,测定结果的相对标准偏差为0.43%~0.81%(n=6)。该方法简单可靠,重复性好,适用于异丙醇-水溶液中利血平含量的测定。A method for the determination of reserpine in isopropyl alcohol-aqueous solution by high performance liquid chromatography was established.Separation was performed on Eclipse Plus C18 column(150 mm×4.6 mm,5μm).Equal elution was performed by flow equielution with a volume ratio of methanol to aqueous solution of 8∶2.A diode array detector with wavelength of 268 nm,flow rate of 0.5 mL/min and column temperature of 40℃were used for detection.The mass concentration of reserpine were in the range of 0.2-10μg/mL,the correlation coefficient was 0.9999,and the detection limit was 0.05μg/mL.The recoveries were 100.20%-100.39%,and the relative standard deviations were 0.43%-0.81%(n=6).The method is simple,reliable and reproducible,which can be used for the determination of reserpine in isopropyl alcohol-aqueous solution.
