摘  要:以细叶石斛(Dendrobium hancockii Rolfe)成熟种子为外植体,研究不同激素组合对原球茎途径的增殖、分化以及壮苗生根的影响。结果表明:细叶石斛种子萌发培养50 d即可出现大量饱满绿色的原球茎;增殖、分化阶段最佳培养基为MS培养基+1.0 mg·L^(–1)6-苄氨基嘌呤(6-BA)+0.3 mg·L^(–1)萘乙酸(NAA)+蔗糖30 g·L^(–1)+琼脂7.8 g·L^(–1),增殖率为16.67%;生根壮苗最适培养基为MS(Murashige-Skoog)培养基+0.5 mg·L^(–1) NAA+0.1 mg·L^(–1)吲哚丁酸(indolebutyric acid,IBA),生根率100%,分化系数为6.14,幼苗健壮且生长快速。优化了细叶石斛组培快繁体系,为后续进一步优化以及工厂化生产提供依据。Utilizing the mature seeds of Dendrobium hancockii Rolfe as explants,this study investigated the impacts of various hormone combinations on the proliferation,differentiation,and rooting of protocorm-like bodies(PLBs).The results indicated that a large number of plump and green PLBs emerged from the Dendrobium hancockii Rolfe seeds after 50 days of germination cultivation.The optimal medium for proliferation and differentiation was MS(Murashige-Skoog)medium supplemented with+1.0 mg·L^(–1)6-BA+0.3 mg·L^(–1) NAA+30 g·L^(–1) sucrose+7.8 g·L^(–1) agar,with a proliferation rate of 16.67%.The optimal medium for rooting and seedling growth was MS medium+0.5 mg·L^(–1) NAA+0.1 mg·L^(–1) IBA,with a rooting rate of 100%and a differentiation coefficient of 6.14,resulting in robust seedlings with rapid growth.This study optimized the tissue culture and rapid propagation system of Dendrobium hancockii Rolfe,providing a basis for further optimization and industrial production.
