摘 要:目的:通过胺碘酮诱导静脉炎,观察异甘草素对受损的人脐静脉内皮细胞的保护作用。方法:将人脐静脉内皮细胞分为空白对照组、胺碘酮组(胺碘酮30μmol/L处理24 h)、异甘草素组(异甘草素10μmol/L预处理1 h后胺碘酮30μmol/L处理24 h)3组。采用细胞计数试剂(CCK-8)法确定异甘草素作用浓度,划痕实验观察细胞迁移能力,流式细胞术检测细胞凋亡程度,聚合酶链式反应(PCR)检测增殖因子增殖细胞核抗原(PCNA)、凋亡因子Casepase-3、炎症因子肿瘤坏死因子(TNF-α)、白细胞介素-6(IL-6)、黏附因子细胞间黏附分子-1(ICAM-1)和血管细胞黏附因子-1(VCAM-1)的表达;蛋白质免疫印迹法检测黏附因子ICAM-1和VCAM-1蛋白表达。结果:与胺碘酮组比较,异甘草素预处理可促进细胞增殖和迁移,抑制细胞凋亡,下调相关炎症因子表达。结论:异甘草素在预防和治疗临床静脉炎方面有很好的应用前景,可作为一种潜在的预防静脉炎的治疗药物。Objective:The aim of this study was to observe the protective effect of isoliquiritigenin(ISL)on damaged human umbilical vein endothelial cells(HUVECs)induced by amiodarone(AM)-induced venous inflammation.Methods:HUVECs were divided into three groups:control group,AM group(treated with AM at a concentration of 30μmol/L for 24 h),and ISL group(pretreated with ISL at a concentration of 10μmol/L for 1 h followed by AM at a concentration of 30μmol/L treatment for 24 h).The CCK-8 assay was used to determine the effective concentration of ISL.The scratch assay was performed to evaluate cell migration ability.Flow cytometry was used to assess cell apoptosis.Polymerase chain reaction(PCR)was conducted to detect the expression of proliferation factor PCNA and apoptotic factor Casepase-3.The expression of inflammatory factors tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6),and adhesion factors intercellular cell adhesion molecule-1(ICAM-1)and vascular cell adhesion molecule-1(VCAM-1)were measured.Protein levels of ICAM-1 and VCAM-1 were detected using Western Blot.Results:Compared with the AM group,ISL pretreatment promoted cell proliferation and migration,inhibited cell apoptosis,and downregulated related inflammatory factors.Conclusion:ISL shows promising application prospects in preventing and treating clinical venous inflammation,and can be considered as a potential therapeutic drug for preventing venous inflammation.
