丹荷颗粒对雄性/雌性混合型高脂血症大鼠降脂作用初探及比较

摘　　要：目的探讨丹荷颗粒对雄性/雌性混合型高脂血症大鼠的降脂作用。方法将60只大鼠(雌雄各半)随机分为正常对照组和造模组,正常对照组大鼠给予普通饲料喂养,造模组给予高脂饲料连续喂养,2周后,建立大鼠混合型高脂血症模型。造模成功后,按大鼠总胆固醇(total cholesterol,TC)值随机分为模型组,丹荷颗粒低、中、高剂量组,普伐他汀钠组,每组5只。造模第3周开始予相应药物灌胃,连续6周。称量大鼠体质量,称取肝湿重,计算肝指数,试剂盒检测TC、甘油三酯(total triglycerides,TG)、低密度脂蛋白胆固醇(low density lipoprotein cholesterol,LDL-C)、高密度脂蛋白胆固醇(high density lipoprotein cholesterol,HDL-C),HE染色观察肝组织病理变化。结果与正常对照组比较,两种性别模型组大鼠体质量增长,血清TG水平升高(P<0.05,P<0.01),有不同程度肝脂肪变性和肝损伤,且雄性高于雌性,血清TC、LDL-C水平升高,HDL-C水平降低(P<0.05,P<0.01),且雌性高于雄性;与模型组相比,丹荷颗粒各剂量组显著降低了两种性别大鼠血清TC、TG、LDL-C水平(P<0.05,P<0.01);丹荷颗粒显著升高了雌性大鼠HDL-C水平(P<0.01),对雄性大鼠无影响;与正常对照组比较,丹荷颗粒各剂量组减轻了两种性别大鼠的肝脂肪变性及肝损伤。结论丹荷颗粒对混合型高脂血症两种性别大鼠具有降脂作用,可有效降低肝脂肪变性和肝损伤,其效果雄性优于雌性。Objective To investigate the hypolipidemic effect of Danhe Granules(丹荷颗粒)on male/female mixed hyperlipidemia rats.Methods 60 rats(half male and half female)were randomly divided into normal control group and modeling group.The rats in normal control group were fed with normal chow,and those in modeling group were fed with high-fat chow continuously for 2 weeks to establish a mixed hyperlipidemia model in rats.After successful modeling,the rats were randomly divided into the model group,the low,medium and high dose groups of Danhe Granules,and the pravastatin sodium group according to their the total cholesterol(TC)values,with 5 rats in each group.The corresponding drugs were given by gavage from the 3rd week of modeling for 6 weeks.The body mass of the rats was weighed,the wet weight of the liver was weighed,the hepatic index was calculated,and TC,triglyceride(TG),low density lipoprotein cholesterol(LDL-C),and high density lipoprotein cholesterol(HDL-C)were detected by the kit,and the pathological changes of the liver histology were observed by HE staining.Results Compared with the normal control group,rats in the model group of both genders showed increased body mass,elevated serum TG(P<0.05,P<0.01),different degrees of hepatic steatosis and liver injury,and males were higher than females,elevated serum TC and LDL-C,and decreased HDL-C(P<0.05,P<0.01),and females were higher than males;compared with the model group,the Danhe Granules in each of the dosed groups significantly reduced the liver histopathological changes.group significantly reduced serum TC,TG and LDL-C levels in rats of both genders(P<0.05,P<0.01);Danhe Granules significantly increased HDL-C levels in females(P<0.01)and had no effect on males;and compared with the normal control group,Danhe Granules of each dosage group attenuated hepatic steatosis and liver injury in rats of both genders.Conclusion Danhe Granules have lipid-lowering effect on rats of both sexes with mixed hyperlipidemia,and can effectively reduce hepatic steatosis and live
