围刺对黄褐斑豚鼠皮肤氧化应激及黑色素合成影响

摘  要:目的探讨围刺对黄褐斑(melasma)豚鼠模型皮肤的影响及作用机制。方法32只普通级雌性豚鼠随机分为空白组(8只)和造模组(24只),造模组采用中波紫外线照射配合黄体酮注射诱导黄褐斑模型,建模30 d后,根据表观观察,苏木素(HE)、免疫组化染色法观察皮肤组织病理变化,ELISA法检测组织中超氧化物歧化酶(superoxide dismutase,SOD)活性、丙二醛(malondialdehyde,MDA)含量,检验模型是否制备成功。造模成功后随机分为模型组、西药组、针药组,每组各8只。空白组和模型组只进行相同条件下的抓取,不予其他干预;西药组予以5%氨甲环酸乳膏外用0.5 g,2次/d;针药组在外用西药的同时予以皮损区围刺,留针30 min,1次/d,连续干预28 d。对各组豚鼠皮肤进行表观指标评价,HE、免疫组化染色法分别观察各组皮肤组织形态学及黑色素细胞的数目变化,ELISA法检测各组皮肤组织中SOD活性,MDA、小眼畸形相关转录因子(microphthalmia-related transcription factor,MITF)及酪氨酸酶(tyrosinase,TYR)的含量表达情况。结果造模后,与空白组对比,造模组皮肤显现出红褐色斑块,伴有脱屑结痂;皮肤表皮层及真皮层结构明显紊乱;基底层及棘层内可见数量较多的阳性黑色素细胞(melanocytes,MC);皮肤组织中SOD活性降低,MDA含量升高(P<0.01),提示黄褐斑豚鼠模型制备成功。干预后,与模型组相比,西药组、针药组皮肤表观指标均有好转,但针药组更为明显;与模型组相比,西药组、针药组皮肤病理结构均有改善,而针药组尤为突出;与模型组相比,西药组、针药组皮肤中阳性MC个数、数密度均明显下降(P<0.05,P<0.01),以针药组最为显著(P<0.05);与模型组相比,西药组、针药组皮肤组织中SOD活性均明显升高,MDA、MITF、TYR含量均明显降低(P<0.05,P<0.01),以针药组更为明显(P<0.05)。结论利用中波紫外线照射配合黄体酮注射可成功制备黄褐斑豚鼠模型,围刺可Objective To investigate the effect and mechanism of perispinal spines on the skin of guinea pig models of melasma.Methods Thirty-two female guinea pigs were randomly divided into blank group(n=8)and module(n=24),the model was induced by ultraviolet irradiation combined with progesterone injection,and after 30 days of modeling,the pathological changes of skin tissue were observed by HE and immunohistochemical staining,and the activity of superoxide dismutase(SOD)and malondialdehyde(MDA)content in tissues were detected by ELISA to test whether the model was successfully prepared.After successful modeling,they were randomly divided into model group,Western medicine group,and acupuncture group,With 8 animals in each group.The blank group and the model group were only subjected to grasping under the same conditions,and no other intervention was given,and the western medicine group was given 5%tranexamic acid cream 0.5 g twice a day,and the acupuncture group was given peripuncture of the skin lesion area while using the western medicine externally,and the needle was left for 30 minutes,once a day,for 28 days.Evaluation of epigenetic indicators was performed on the skin of guinea pigs in each group.HE and immunohistochemistry staining methods were used to observe the morphological changes of skin tissue and the number of melanocytes in each group.ELISA was used to detect the activity of SOD and the expression of MDA,microphthalmia-related transcription factor(MITF)and tyrosinase(TYR)in skin tissues in each group.Results After modeling,compared with the blank group,the skin of the mold showed reddish-brown patches with desquamation and crusting,the structure of the epidermis and dermis layer of the skin was obviously disordered,a large number of positive melanocytes(MC)were seen in the basal layer and acanthus,and the activity of SOD in skin tissues was decreased and the content of MDA increased(P<0.01),indicating that the Chloasma guinea pig model was successfully prepared.After treatment,compared with the model group,
