独活寄生汤抑制铁死亡治疗膝骨关节炎研究进展

摘　　要：膝骨关节炎(knee osteoarthritis,KOA)是常见的骨关节退行性病变,特征为软骨退变、软骨下骨硬化和滑膜炎症,发病机制复杂。铁死亡通过脂质过氧化、炎症及氧化应激相互作用,导致软骨退化、滑膜炎症等,加速KOA进程。文章从铁超载、氨基酸代谢异常、脂质代谢异常等3个方面归纳总结铁死亡参与基质金属蛋白酶13(matrix metalloproteinase 13,MMP-13)的表达、谷氨酸的代谢、脂质过氧化在KOA中的作用和相关机制。该文通过检索PubMed、CNKI、WanFang等数据库近15年来关于独活寄生汤治疗KOA的文献,旨在综述其药理作用和机制,并分析独活寄生汤中的有效成分,综述独活寄生汤对抑制铁死亡作用研究进展,分析独活寄生汤在KOA疾病中的治疗潜力,为中医药治疗KOA提供新思路。Knee osteoarthritis(KOA)is a common degenerative joint disease characterized by cartilage degeneration,subchondral bone sclerosis,and synovial inflammation,with a complex pathogenesis.Ferroptosis,through interactions involving lipid peroxidation,inflammation,and oxidative stress,leads to cartilage degradation,synovial inflammation,and accelerates the progression of KOA.This article summarizes the involvement of ferroptosis in KOA through iron overload,abnormal amino acid metabolism,and abnormal lipid metabolism,focusing on the expression of matrix metalloproteinase 13(MMP-13),glutamate metabolism,lipid peroxidation,and related mechanisms in KOA.This article aims to review the pharmacological effects and mechanisms of Duhuo Jisheng Decoction(独活寄生汤)in the treatment of KOA by searching PubMed,WOSCC,CNKI,WanFang and other databases in the past 15 years.It also analyzes the active ingredients of Duhuo Jisheng Decoction and reviews the research progress on its inhibitory effect on ferroptosis,assessing the therapeutic potential of Duhuo Jisheng Decoction in KOA and providing new insights for traditional Chinese medicine treatment of KOA.
