五虎汤通过干预P53/NF-κB串扰改善幼龄哮喘小鼠气道炎症机制研究

摘  要:目的:探讨五虎汤通过干预P53/NF-κB串扰对幼龄哮喘小鼠气道炎症的影响。方法:SPF级幼龄(3周龄)雄性BALB/c小鼠60只,随机将10只小鼠作为空白组,50只作为造模组。采用RSV复合鸡卵清蛋白(OVA)雾化吸入诱发哮喘小鼠模型,造模成功后,随机分为模型组、地塞米松组和五虎汤低、中、高剂量组,每组10只。分组干预后,每日记录行为学改变。干预2周后,HE染色法观察小鼠肺组织气道炎症细胞浸润;ELISA检测血清肿瘤坏死因子-α(TNF-α)以及白介素-6(IL-6)的含量;Western blot法检测肺组织P53和P65蛋白表达,并对P53与P65蛋白含量进行相关性分析。结果:与空白组比较,模型组小鼠形体较小,饮食不佳,容易激惹,撕咬增多,多动搔抓,点头样呼吸,喷嚏喘息,呼吸加快,提示造模成功。与空白组比较,模型组小鼠肺组织周围炎性细胞浸润;血清IL-6、TNF-α含量呈现显著上升趋势(均P<0.05);肺组织P53与P65蛋白表达呈显著上升趋势(均P<0.05)。与模型组相比,五虎汤低、中、高剂量组及地塞米松组小鼠行为学改善明显,肺组织炎性细胞浸润明显减少,且五虎汤高剂量组与地塞米松组改善更加明显;血清IL-6、TNF-α含量明显降低(均P<0.05);肺组织P53和P65蛋白表达明显下降(均P<0.05)。Pearson相关性分析显示,小鼠肺组织P53与P65有极强正相关性(r=0.925,P<0.05)。结论:五虎汤能够改善哮喘小鼠的气道炎症,其机制可能与降低P53、P65蛋白表达,抑制NF-κB的转录有关。Objective:Discuss the effects of Wuhu decoction through intervention in P53/NF-κB skewers on the inflammation of young asthma mice.Methods:There were 60 SPF grade young(3 weeks old)male BALB/c mice,10 mice were randomly selected as the blank group and 50 mice as the modeling group.Using the RSV composite chicken ovard protein(OVA)atomization inhalation to induce asthma mouse models.After successful modeling,they were randomly divided into model group,dexamethasone group and Wuhu decoction low-dose,medium-dose and high-dose groups,with 10 animals in each group.Behavioral changes were recorded daily after group intervention.After two weeks of intervention,airway inflammatory cell infiltration in lung tissue was observed by HE staining.Serum tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)were detected by ELISA.Western blot was used to detect the expression of P53 and P65 protein in lung tissue,and the correlation between P53 and P65 protein content was analyzed.Results:Compared with the blank group,the mice in the model group were smaller,had poor diet,were easily irritated,more biting,more scratching,nodding breathing,sneezing and wheezing,and breathing quickened,indicating that the modeling was successful.Compared with blank group,inflammatory cells were infiltrated around lung tissue in model group.The contents of IL-6 and TNF-αin serum were significantly increased(all P<0.05).The expression of P53 and P65 protein in lung tissue was significantly increased(all P<0.05).Compared with the model group,the behavior of mice in Wuhu decoction low,medium and high dose-groups and dexamethasone group was significantly improved,and the inflammatory cell infiltration of lung tissue was significantly reduced,and the improvement was more obvious in Wuhu decoction high-dose group and dexamethasone group.Serum IL-6 and TNF-αcontents were significantly decreased(all P<0.05).The protein expressions of P53 and P65 in lung tissue were significantly decreased(all P<0.05).Pearson correlation analysis showed that there was a
