摘 要:【目的】研究草分枝杆菌短时(进食后4 h)对断奶仔猪肠道微生物群落的影响。【方法】通过体外静置培养实验,将断奶仔猪进食后4 h盲肠内容物用生理盐水稀释(1∶10)后作为接种物,以1∶9的比例将接种物接种至培养基。以添加10μg/mL的草分枝杆菌干粉作为处理组,以等量无菌石英砂作为对照组。对照组和试验组均置于37℃二氧化碳培养箱中培养4 h,在12000 r/min冷冻离心条件下离心10 min,并用16S RNA基因测序技术测定其微生物组成。【结果】实验结果显示:相较于对照组,添加草分枝杆菌制剂的断奶仔猪盲肠内容物(培养后4 h)中,Oribacterium、梭状芽孢杆菌属(Clostridium sensu stricto)、大肠埃希氏菌(Escherichia coli)等细菌数量显著减少;而罗伊氏乳杆菌(Lactobacillus reuteri)、唾液乳杆菌(Lactobacillus salivarius)、戊糖片球菌(Pediococcus pentosaceus)、魏斯氏菌属(Weissella)等细菌数量则显著增加。【结论】草分枝杆菌制剂在短时间内能够有效调节断奶仔猪肠道微生物的丰度,显示出其对肠道微生物群落的积极影响。【Objective】This study aimed to investigate the short-term regulatory effects of Mycobacterium phlei preparation on the cecal microbiota of weaned piglets through an in vitro static culture experiment,analyzing the impact of M.phlei on the intestinal microbial community.【Method】Cecal contents from weaned piglets were diluted with physiological saline(1∶10)and used as the inoculum,which was inoculated into the medium at a 10%volume.The treatment group was supplemented with 10μg/mL of M.phlei powder,while an equal volume of sterile quartz sand served as the control group.Both the control and treatment groups were incubated in a 37℃carbon dioxide incubator for 4 h.After incubation,samples were centrifuged at 12000 r/min for 10 min,and the microbial composition was determined using 16S RNA technology.【Result】The experimental results showed that the cecal microbiota of weaned piglets was primarily composed of the phyla Firmicutes,Bacteroidetes,and Proteobacteria.Among the top ten genera in terms of microbial abundance were Alloprevotella,Prevotella,Lactobacillus,and Oribacterium.Compared to the control group,the treatment group exhibited a significant reduction in the number of bacteria such as Oribacterium,Clostridium sensu stricto,and Escherichia coli.Conversely,there was a significant increase in the number of bacteria such as Lactobacillus reuteri,Lactobacillus salivarius,Pediococcus pentosaceus,and Weissella.【Conclusion】In summary,the M.phlei preparation effectively modulated the abundance of intestinal microbiota in weaned piglets within a short period,demonstrating its positive impact on the intestinal microbial community.This finding provides strong experimental evidence for the application of M.phlei in animal husbandry and lays a foundation for further investigation into its mechanism of action and potential applications.
