摘 要:了解染病柠檬叶片中内生细菌的种类和多样性,并从中筛选出对柑橘溃疡病菌有拮抗作用的内生菌进行鉴定。以感染柑橘溃疡病的柠檬病叶为材料,利用稀释分离法对病叶中可培养的内生细菌进行分离、纯化;结合菌落形态特征和16SrDNA序列对所获得内生细菌进行初步鉴定;随后采用平板对峙法测定所分离的内生细菌对柑橘溃疡病菌的拮抗作用。结果表明,从病叶中共分离到可培养细菌42株,分为13种不同类型,变形菌门检出的种类最多,占总数的61.9%,放线菌门其次,占总数的28.6%,厚壁菌门最少,占内生菌总数的9.5%。拮抗活性测定表明,菌株nx28、nx37对柑橘溃疡病菌有抑制作用,平均抑菌圈直径为37.6 mm,经鉴定菌株nx28、nx37均为解淀粉芽孢杆菌。The purpose of this study was to investigate the diversity of endophytic bacteria in lemon leaves affected by citrus canker and identify the antagonistic strains against Xanthomonas citri subsp.Citri(Xcc).The culturable endophytic bacteria were isolated and purified from infected lemon leaves via the gradient plate dilution method and then identified based on morphology combined with the 16S rDNA sequence.The antagonism of isolated endophytic bacteria against Xcc was detected via the plate confrontation method.The results showed that 42 cultivable bacteria were isolated from lemon leaves affected by citrus canker and divided into 13 types.Proteobacteria was the most domain phyla,accounting for more than 61%of the total number of endophytic bacteria,followed by Actinobacteria,accounting for 28.6%,and Firmicutes was the least,accounting for 9.5%.The determination of antagonistic activity revealed that the strains nx28 and nx37 had inhibitory effects on the pathogen of citrus canker with an average inhibitory circle diameter of 37.6 mm.Both strains nx28 and nx37 were identified as Bacillus amyloliquefaciens.
