摘  要:目的观察脂联素受体激动剂AdipoRon对糖尿病小鼠睾丸组织中内质网应激及细胞焦亡表达水平的影响,初步探讨AdipoRon保护糖尿病小鼠睾丸的作用机制。方法8周龄雄性C57BL/6小鼠随机分为对照组(NC组)、糖尿病组(DM组)、AdipoRon治疗组(ADI组)。采用链脲佐菌素(STZ)腹腔注射制备糖尿病小鼠模型。造模成功后,ADI组给予AdipoRon灌胃(20 mg/kg),NC组和DM组分别给予相同剂量的生理盐水灌胃。药物干预治疗8周后,采用酶联免疫吸附法(ELISA)检测空腹血糖、胰岛素、睾酮、卵泡刺激素及黄体生成素,测量睾丸质量并计算睾丸指数,检测精子数量及活动度,HE染色观察睾丸组织形态变化。荧光定量PCR法测定睾丸组织中内质网应激指标(GRP78、CHOP、Caspase-12)和焦亡相关蛋白(GSDMD)mRNA表达量。结果与NC组相比,DM组小鼠睾丸生精小管数目减少,生精细胞数量减少且排列疏松,血清胰岛素、睾酮、黄体生成素、卵泡刺激素、精子活动率、精子数量水平均显著降低(P<0.05),空腹血糖和睾丸质量指数明显上调(P<0.05),且睾丸组织中GRP78、CHOP、Caspase-12、GSDMD的mRNA表达水平均显著升高(P<0.05)。与DM组相比,ADI组小鼠睾丸病理损伤有所减轻,血清胰岛素、睾酮、黄体生成素、体质量、精子活动率及精子数量水平显著升高(P<0.05),血糖、睾丸指数及睾丸组织中GRP78、CHOP、Caspase-12、GSDMD的mRNA表达水平显著降低(P<0.05)。结论AdipoRon可能通过调控内质网应激及细胞焦亡对糖尿病小鼠睾丸组织产生保护作用。Objective To investigate the effects of AdipoRon on the expression levels of endoplasmic reticulum stress and pyroptosis in testis tissues of diabetic mice,and explore the mechanism of AdipoRon protecting the testis of diabetic mice.Methods Eight-week-old male C57BL/6 mice were randomly divided into normal control group(NC group),diabetes group(DM group)and AdipoRon treatment group(ADI group).The diabetic mouse model was prepared by intraperitoneal injection of streptozotocin(STZ).After suc-cessful modeling,the mice were given AdipoRon(20 mg/kg)by gavage in ADI group,and the same dose of saline in NC group and DM group.After eight weeks of drug intervention,the levels of fasting blood glucose,insulin,testosterone,follicle stimulating hor-mone and luteinizing hormone were measured by enzyme-linked immunosorbent assay(ELISA).Testicular mass was measured,testicu-lar index was calculated,and sperm number and activity were measured.Morphological changes of testis tissue were observed using he-matoxylin-eosin(HE)staining.The mRNA expressions of endoplasmic reticulum stress-related proteins(GRP78,CHOP,Caspase-12)and pyroptosis-related protein(GSDMD)in testis tissue were measured using fluorescence quantitative PCR.Results Compared with NC group,the number of seminiferous tubules in the testis and the number of spermatogenic cells decreased in DM group,and the arrangement of spermatogenic cells was loose,the levels of serum insulin,testosterone,luteinizing hormone,follicle-stimulating hormone,sperm motility and sperm counts were significantly reduced(P<0.05),fasting blood glucose and testicular mass index were significantly increased(P<0.05),the expressions of GRP78,CHOP,Caspase-12 and GSDMD mRNA in testis tissue were signifi-cantly increased(P<0.05).Compared with DM group,the testicular pathologic damage was attenuated in ADI group,the levels of se-rum insulin,testosterone,luteinizing hormone,body weight,sperm motility,and sperm count were significantly increased(P<0.05),and the levels of fasting blood glucose,testicula
